Hier presenteren we een protocol om een genetisch gelabelde subpopulatie van neuronen door een twee-foton laser vanaf Zebrafish larven lasertherapie.
Ter identificatie van de rol van een subpopulatie van neuronen in gedrag, is het essentieel om te testen van de gevolgen van het blokkeren van de activiteit in levende dieren. Laser ablatie van neuronen is een effectieve methode voor dit doel als neuronen zijn selectief gelabeld met de fluorescerende sondes. In de huidige studie, worden protocollen voor laser geruineerd een subpopulatie van neuronen met behulp van een twee-foton microscoop en het testen van de functionele en gedragsmatige gevolgen daarvan beschreven. In deze studie, wordt prooi vangen gedrag in zebrafish larven gebruikt als een studie-model. Het pretecto-hypothalame circuit is bekend dat ten grondslag liggen aan deze visueel gestuurde prooi vangen van gedrag. Zebravis pretectum waren laser-ablated worden, en neuronale activiteit in de inferieure kwab van de hypothalamus (ILH; het doel van de pretectal projectie) werd onderzocht. Prooi vangen gedrag na pretectal ablatie werd ook getest.
Om te begrijpen hoe gedrag vloeit voort uit de neuronale activiteit in de hersenen, is het noodzakelijk om te identificeren van de neurale circuits die bij de generatie van dat gedrag betrokken zijn. Het larvale stadium, de zebravis biedt een ideale dierlijk model voor het bestuderen van de hersenfunctie geassocieerd met het gedrag, omdat hun kleine, transparante hersenen maken het mogelijk om te onderzoeken van neuronale activiteit op een cellulaire resolutie op een breed gebied van de hersenen met inachtneming van de gedrag1. Beeldvorming van neuronale activiteit in specifieke neuronen is mogelijk door de uitvinding van genetisch gecodeerde calcium (Ca) indicatoren (GECIs) zoals GCaMP2geworden. GCaMP transgene zebrafish hebben bewezen nuttig voor het koppelen van het functionele neurale circuit met gedrag door het uitvoeren van Ca imaging in dieren3gedragen.
Terwijl Ca beeldvorming kan aantonen dat de correlaties tussen Neuronale activiteit en gedrag, om aan te tonen van causaliteit, onderdrukking van neuronale activiteit en haar gevolgen op gedrag testen zijn belangrijke stappen. Er zijn verschillende manieren om dit te bereiken: het gebruik van genetische mutatie die wijzigt van specifieke neurale circuits4, uitdrukking van neurotoxines in specifieke neuronen5,6, gebruik van optogenetic hulpmiddelen zoals halorhodopsin7, en laser ablatie van gerichte neuronen8,9. Laser Ablatie is bijzonder geschikt voor het elimineren van de activiteit in een relatief klein aantal specifieke neuronen. Onomkeerbare eliminatie van neuronale activiteit door het doden van neuronen vergemakkelijkt gedragsmatige gevolgen.
Een interessant gedrag dat kan worden waargenomen in het larvale stadium van zebravis is prooi vangen(Figuur 1). Deze visueel geleide, doelgericht gedrag biedt een gunstige experimenteel systeem voor de studie van de gezichtsscherpte10, visuomotor transformatie11,12,13, visuele perceptie en erkenning van objecten14,15,16,17,18jp19van de besluitvorming. Hoe prooi wordt herkend door roofdieren en hoe prooi detectie leidt tot prooi vangen van gedrag is een centrale vraag in neuroethology20. In dit artikel focussen we op de rol van het pretecto-hypothalame circuit gevormd door projecties van een kern in de pretectum (nucleus pretectalis superficialis pars magnocellularis, hierna gewoon bekend als de pretectum) aan de ILH. Laser-ablatie van de pretectum bleek te verminderen prooi vangen activiteit en neuronale activiteit in de ILH die is gekoppeld aan de visuele prooi perceptie21af te schaffen. Hier, laser protocollen voor het uitvoeren van ablatie en testen zijn effect met behulp van Ca2 + beeldvorming en gedrags opname in zebrafish larven worden beschreven.
Hoewel de twee-foton laser een uitstekende ruimtelijke resolutie heeft te specifiek lasertherapie individuele neuronen, moet grote voorzichtigheid worden genomen om te voorkomen dat ongewenste schade op het hersenweefsel als gevolg van warmte. De belangrijkste stap in het baarmoederslijmvlies experiment is om te bepalen van de optimale hoeveelheid laser-bestraling. Onvoldoende bestraling mislukt om te doden van de neuronen. Te veel bestraling zal warmte-schade het omringende weefsel, die zal resulteren in ongewenste effe…
