De vernietiging van specifieke cellen in het embryo is een krachtig hulpmiddel voor het bestuderen van cellulaire interacties die betrokken zijn bij het lot van cellen. Het huidige protocol beschrijft technieken voor de laserablatie van gerichte cellen in het vroege embryo van de bruine alg Saccharina latissima.
In Saccharina latissima ontwikkelt het embryo zich als een enkellaags celblad dat de lamina of het mes wordt genoemd. Elke embryocel is gemakkelijk waar te nemen, gemakkelijk te onderscheiden van zijn buren en kan individueel worden gericht. Al tientallen jaren wordt laserablatie gebruikt om de ontwikkeling van embryo’s te bestuderen. Hier werd een protocol voor celspecifieke laserablatie ontwikkeld voor vroege embryo’s van de bruine alg S. latissima. Het gepresenteerde werk omvat: (1) de voorbereiding van Saccharina-embryo’s , met een beschrijving van de kritische parameters, inclusief kweekomstandigheden, (2) de laserablatie-instellingen en (3) de monitoring van de daaropvolgende groei van het bestraalde embryo met behulp van time-lapse-microscopie. Daarnaast worden details gegeven over de optimale omstandigheden voor het transport van de embryo’s van het beeldvormingsplatform terug naar het laboratorium, wat de latere embryo-ontwikkeling diepgaand kan beïnvloeden. Algen van de orde Laminariales vertonen embryogenesepatronen die vergelijkbaar zijn met Saccharina; dit protocol kan dus gemakkelijk worden overgedragen op andere soorten in dit taxon.
Laserablatie wordt al tientallen jaren gebruikt om de ontwikkeling van embryo’s te bestuderen. Het bestralen van embryocellen met een laserstraal maakt het mogelijk om het regeneratieve potentieel en de wijziging van de cellijn tijdens de embryogenese te monitoren en de impact van gerichte ablatie op de celdeling en het lot van de cel te onderzoeken. De modelorganismen die worden gebruikt in laserablatiemethoden zijn meestal dieren, zoals insecten 1,2, nematoden 3,4, gewervelde dieren 5,6 en af en toe planten 7,8. Daarnaast werd in 1994 en 1998 een lasermicro-ablatiebenadering gebruikt op de bruine alg Fucus om de rol van de celwand in de fotopolarisatie van het vroege embryo aan te tonen 9,10.
Bruine algen behoren tot de groep Stramenopiles, die 1,6 miljard jaar geleden aan de wortel van de eukaryote boom uiteenvielen. Als gevolg hiervan zijn ze fylogenetisch onafhankelijk van andere meercellige organismen, zoals dieren en planten11. Saccharina latissima behoort tot de orde Laminariales, beter bekend als kelps, en ze behoren tot de grootste organismen op aarde, met afmetingen van meer dan 30 m. Saccharina sp. is een groot zeewier dat wordt gebruikt voor vele toepassingen zoals voedsel en diervoeder, en de polysacchariden worden geëxtraheerd voor gebruik in de landbouw-, farmacologische en cosmetische industrie wereldwijd12, 13. De teelt ervan, voornamelijk in Azië en meer recent in Europa, vereist de voorbereiding van embryo’s in broederijen voordat juvenielen in de open zee worden vrijgelaten. Zoals alle kelps heeft het een bifasische levenscyclus die bestaat uit een microscopische gametofytische fase, waarin een haploïde gametofyt groeit en gameten produceert voor bevruchting, en een diploïde macroscopische sporofytische fase, waarbij een groot planair blad zich ontwikkelt uit zijn holdfast bevestigd aan de zeebodem of rotsen. De sporofyt geeft op volwassen leeftijd haploïde sporen af, waardoor de levenscyclus 14,15,16 wordt voltooid.
S. latissima presenteert enkele interessante morfologische kenmerken17. Het embryo ontwikkelt zich als een enkellaags planair vel 15,18,19 voordat het een meerlagige structuur krijgt die samenvalt met de opkomst van verschillende weefseltypen. Bovendien is Laminariales een van de weinige taxa van bruine algen waarvan de embryo’s gehecht blijven aan hun maternale gametofytische weefsel (Desmarestiales en Sporochnales doen dat ook15). Deze functie biedt de mogelijkheid om de rol van moederlijk weefsel in dit ontwikkelingsproces te bestuderen en maternale controlemechanismen in bruine algen te vergelijken met die in dieren en planten.
Dit artikel presenteert het eerste complete protocol voor laserablatie in een vroeg kelp-embryo. Dit protocol met UV ns-gepulseerde techniek resulteert in de specifieke vernietiging van individuele embryocellen om hun respectieve rollen tijdens embryogenese te bestuderen. De procedure biedt een betrouwbare aanpak voor het onderzoeken van celinteracties en cellotgevallen tijdens embryogenese in Laminariales.
Lokale cellulaire laserablatie maakt temporele en ruimtelijke ablatie mogelijk met een hoge mate van precisie. De efficiëntie ervan kan echter worden belemmerd door de niet-toegankelijkheid van doelcellen; alle cellen zijn bijvoorbeeld van een driedimensionaal embryo. Dit protocol is ontwikkeld op het embryo van de alg Saccharina latissima, dat een monolayered lamina ontwikkelt waarin alle cellen gemakkelijk kunnen worden onderscheiden en individueel kunnen worden vernietigd met een laserstraal.
