Dit artikel beschrijft de isolatie en het kweken van embryonale rat sympathische neuronen van de superieure cervicale ganglia. Het biedt ook gedetailleerde protocollen voor immunocytochemische kleuring en voor de voorbereiding van neuronale extracten voor massaspectrometrische analyse.
Sympathische neuronen van de embryonale rat superieure cervicale ganglia (SCG) zijn gebruikt als een in vitro model systeem voor perifere neuronen om axonale groei, axonale handel, synaptogenese, dendritische groei, dendritische plasticiteit en zenuw-doel interacties in co-cultuur systemen te bestuderen. Dit protocol beschrijft de isolatie en dissociatie van neuronen van de superieure cervicale ganglia van E21 rat embryo’s, gevolgd door de voorbereiding en het onderhoud van zuivere neuronale culturen in serum-vrij medium. Aangezien neuronen zich niet houden aan ongecoat plastic, neuronen zullen worden gekweekt op 12 mm glazen coverslips of 6-well platen bekleed met poly-D-lysine. Na behandeling met een antimitotisch middel (Ara-C, cytosine β-D-arabinofuranoside), genereert dit protocol gezonde neuronale culturen met minder dan 5% niet-neuronale cellen, die meer dan een maand in vitro kunnen worden gehandhaafd. Hoewel embryonale rat SCG neuronen multipolair zijn met 5-8 dendrieten in vivo; onder serumvrije omstandigheden breiden deze neuronen slechts één axon in de cultuur uit en blijven unipolair voor de duur van de cultuur. Echter, deze neuronen kunnen worden geïnduceerd om dendrieten uit te breiden in de aanwezigheid van kelder membraan extract, bot morfogenetische eiwitten (BMPs), of 10% foetale kalf serum. Deze homogene neuronale culturen kunnen worden gebruikt voor immunocytochemische kleuring en voor biochemische studies. Dit artikel beschrijft ook geoptimaliseerd protocol voor immunocytochemische kleuring voor microtubuli geassocieerde eiwit-2 (MAP-2) in deze neuronen en voor de voorbereiding van neuronale extracten voor massaspectrometrie.
Sympathische neuronen afgeleid van embryonale superieure cervicale ganglia (SCG) zijn op grote schaal gebruikt als een primaire neuronale cultuur systeem voor het bestuderen van vele aspecten van de neuronale ontwikkeling, waaronder groeifactor afhankelijkheid, neuron-target interacties, neurotransmitter signalering, axonale groei, dendriet ontwikkeling en plasticiteit, synaptogenese en signalering mechanismen die ten grondslag liggen aan nerve-target / neuron-glia interacties1,2,3,,4,5,6,7,8,9. Ondanks hun kleine omvang (ongeveer 10000 neuronen / ganglia), zijn er drie belangrijke redenen voor de ontwikkeling en uitgebreid gebruik van dit cultuursysteem zijn i) zijn de eerste ganglia in de sympathieke keten, ze zijn groter, en dus gemakkelijker te isoleren, dan de rest van de sympathieke ganglia10; ii) in tegenstelling tot centrale neuronen, de neuronen in de SCG zijn vrij homogeen met alle neuronen worden afgeleid van de neurale kam, met een vergelijkbare grootte, afhankelijkheid van zenuwgroei factor en wordt noch-adrenerge. Dit maakt ze een waardevol model voor morfologische en genomische studies10,11 en iii) deze neuronen kunnen worden gehandhaafd in een gedefinieerd serum-vrij medium met zenuwgroeifactor voor meer dan een maand10,12. Perinatale SCG neuronen zijn op grote schaal gebruikt voor het bestuderen van de mechanismen die ten grondslag liggen aan de initiatie en het onderhoud van dendrieten2. Dit komt vooral omdat, hoewel SCG neuronen hebben een uitgebreide dendritische prieel in vivo, ze niet uit te breiden denndrites in vitro in de afwezigheid van serum, maar kan worden geïnduceerd om denndrites groeien in aanwezigheid van bepaalde groeifactoren zoals bot morfogenetische eiwitten2,12,13.
Dit artikel beschrijft het protocol voor het isoleren en kweken van embryonale rat SCG neuronen. In de afgelopen 50 jaar zijn primaire neuronale culturen van de SCG voornamelijk gebruikt voor morfologische studies met een beperkt aantal studies die de grootschalige genomische of proteomische veranderingen onderzoeken. Dit is voornamelijk te wijten aan kleine weefselgrootte resulterend in de isolatie van lage hoeveelheden DNA of eiwit, waardoor het moeilijk is om genomische en proteomische analyses uit te voeren op deze neuronen. In de afgelopen jaren heeft de toegenomen detectiegevoeligheid echter de ontwikkeling van methoden mogelijk gemaakt om het genoom, miRNome en proteoom in de SCG-neuronen te onderzoeken tijdens de dendritische groeiontwikkeling14,15,16,17. Dit document zal ook de methode beschrijven voor morfologische analyse van deze neuronen met behulp van immunocytochemie en een protocol om neuronale eiwitextracten te verkrijgen voor massaspectrometrische analyse.
Dit artikel beschrijft de protocollen voor het kweken van sympathische neuronen van superieure cervicale ganglia van embryonale rattenpups.
