Alle cijfers in een wetenschappelijke meting zijn zeker, behalve het laatste cijfer. De meetzekerheid hangt af van twee factoren:het aantal cijfers in de meting en de precisie van het gebruikte instrument. In een gemeten grootheid worden alle cijfers, inclusief het laatste onzekere cijfer, significante cijfers genoemd en kunnen ze worden bepaald met behulp van specifieke regels.Alle niet-nul cijfers en alle plaatsopvullende nullen die tussen twee niet-nul cijfers liggen zijn significant. 28 heeft bijvoorbeeld twee significante cijfers, terwijl 26, 25 er vier heeft en 208 er drie heeft. Voorloopnullen zijn nooit significant, ze lokaliseren alleen de komma.0, 00208 heeft bijvoorbeeld drie significante cijfers. Dergelijke hoeveelheden kunnen worden uitgedrukt met behulp van exponentiële notaties. 0, 00208 kan dus worden geschreven als 2.08 10⁻³.Achterliggende nullen zijn alleen significant in decimaal opgemaakte getallen. 2200 heeft twee achterliggende nullen en twee significante cijfers, terwijl 2200, 0 en 2200, 1 beide 5 significante cijfers hebben. Voor hoeveelheden zonder komma wordt de betekenis van achterliggende nullen dubbelzinnig.Zo kan 2200 worden geschreven als 2, 2 10³ met twee significante cijfers of 2, 20 10³ met drie significante cijfers. Significante cijfers helpen ook bij het verkrijgen van zekerheid bij wiskundige bewerkingen. Bij optellen of aftrekken moet het resultaat zo worden afgerond dat het hetzelfde aantal decimalen heeft als de meting met de minste decimalen.Afronding naar beneden moet worden uitgevoerd als het laatste cijfer lager is dan 5, en naar boven moet worden afgerond als het 5 of hoger is. Andere afrondingsmethoden worden soms gebruikt als het laatste cijfer 5 is. Zo wordt de som van 2, 052 en 1, 2 afgerond op 3, 3.Bij het vermenigvuldigen of delen moet het resultaat echter worden afgerond om hetzelfde aantal significante cijfers te hebben als de meting met de minste significante cijfers. Zo wordt het product van 2, 052 en 1, 2 afgerond op 2, 5. Wetenschappers herhalen vaak experimenten om precisie in hun metingen te bereiken.Standaarddeviatie is de statistische uitdrukking van een dergelijke precisie en meet de spreiding ten opzichte van de verwachte waarde. Als de precisie hoog is, is de standaarddeviatie klein en vice versa. Twee groepen maten bijvoorbeeld de dikte van een boek in centimeters.Ze vonden hetzelfde gemiddelde 10, 6 centimeter. De metingen van de eerste groep zijn echter preciezer en hebben dus een lagere standaarddeviatie. De tweede groep heeft meer gespreide metingen en een hogere standaarddeviatie.
