Dit protocol demonstreert de micro-injectie van lipopolysaccharide in het ventriculaire gebied van de hersenen in een zebravislarvemodel om de resulterende neuro-inflammatoire respons en neurotoxiciteit te bestuderen.
Neuro-inflammatie is een belangrijke speler bij verschillende neurologische aandoeningen, waaronder neurodegeneratieve ziekten. Daarom is het van groot belang om alternatieve in vivo neuro-inflammatiemodellen te onderzoeken en te ontwikkelen om de rol van neuro-inflammatie bij neurodegeneratie te begrijpen. In deze studie werd een larvaal zebravismodel van neuro-inflammatie gemedieerd door ventriculaire microinjectie van lipopolysaccharide (LPS) ontwikkeld en gevalideerd. De transgene zebravislijnen elavl3:mCherry, ETvmat2:GFP en mpo:EGFP werden gebruikt voor real-time kwantificering van de levensvatbaarheid van hersenneuronen door fluorescentie live imaging geïntegreerd met fluorescentie intensiteitsanalyse. Het locomotorische gedrag van zebravislarven werd automatisch vastgelegd met behulp van een videotrackingrecorder. Het gehalte aan stikstofmonoxide (NO) en de mRNA-expressieniveaus van inflammatoire cytokines, waaronder interleukine-6 (IL-6), interleukine-1β (IL-1β) en menselijke tumornecrosefactor α (TNF-α) werden onderzocht om de LPS-geïnduceerde immuunrespons in de larvale zebraviskop te beoordelen. Na 24 uur na de hersenventrikelinjectie van LPS werden verlies van neuronen en locomotiedeficiëntie waargenomen bij zebravislarven. Bovendien verhoogde LPS-geïnduceerde neuro-inflammatie de NO-afgifte en de mRNA-expressie van IL-6, IL-1β en TNF-α in het hoofd van 6 dagen na de bevruchting (dpf) zebravislarven, en resulteerde in de rekrutering van neutrofielen in de hersenen van zebravissen. In deze studie werd injectie van zebravissen met LPS in een concentratie van 2,5-5 mg/ml bij 5 dpf bepaald als de optimale conditie voor deze farmacologische neuro-inflammatietest. Dit protocol presenteert een nieuwe, snelle en efficiënte methodologie voor micro-injectie van LPS in de hersenen om LPS-gemedieerde neuro-inflammatie en neurotoxiciteit in een zebravislarve te induceren, wat nuttig is voor het bestuderen van neuro-inflammatie en ook kan worden gebruikt als een high-throughput in vivo screening assay voor geneesmiddelen.
Neuro-inflammatie is beschreven als een cruciale anti-neurogene factor die betrokken is bij de pathogenese van verschillende neurodegeneratieve ziekten van het centrale zenuwstelsel (CZS)1. Na pathologische beledigingen kan neuro-inflammatie leiden tot verschillende nadelige gevolgen, waaronder remming van neurogenese en inductie van neuronale celdood 2,3. In het proces dat ten grondslag ligt aan de respons op ontstekingsinductie, worden meerdere inflammatoire cytokines (zoals TNF-α, IL-1β en IL-6) uitgescheiden in de extracellulaire ruimte en fungeren als cruciale componenten in neurondood en de onderdrukking van neurogenese 4,5,6.
Micro-injectie van ontstekingsmediatoren (zoals IL-1β, L-arginine en endotoxinen) in de hersenen kan neuronale celreductie en neuro-inflammatie veroorzaken 7,8,9. Lipopolysaccharide (LPS, figuur 1), een pathogeen endotoxine dat aanwezig is in de celwand van Gram-negatieve bacteriën, kan neuro-inflammatie induceren, neurodegeneratie verergeren en neurogenese bij dieren verminderen10. LPS-injectie rechtstreeks in het CZS van de muizenhersenen verhoogde niveaus van stikstofmonoxide, pro-inflammatoire cytokines en andere regulatoren11. Bovendien kan stereotaxische injectie van LPS in de lokale hersenomgeving overmatige productie van neurotoxische moleculen induceren, wat resulteert in een verminderde neuronale functie en de daaropvolgende ontwikkeling van neurodegeneratieve ziekten 10,12,13,14,15. Op het gebied van de neurowetenschappen zijn live en tijdsverloop microscopische waarnemingen van cellulaire en biologische processen in levende organismen cruciaal voor het begrijpen van de mechanismen die ten grondslag liggen aan pathogenese en farmacologische werking16. Live beeldvorming van muismodellen van neuro-inflammatie en neurotoxiciteit wordt echter fundamenteel beperkt door de beperkte optische penetratiediepte van microscopie, die functionele beeldvorming en live observatie van ontwikkelingsprocessen uitsluit 17,18,19. Daarom is de ontwikkeling van alternatieve neuro-inflammatiemodellen van groot belang om de studie van pathologische ontwikkeling en het mechanisme dat ten grondslag ligt aan neuro-inflammatie en neurodegeneratie door live beeldvorming te vergemakkelijken.
Zebravis (Danio rerio) is naar voren gekomen als een veelbelovend model om neuro-inflammatie en neurodegeneratie te bestuderen vanwege zijn evolutionair geconserveerde aangeboren immuunsysteem, optische transparantie, grote embryokoppelingsgrootte, genetische tractie en geschiktheid voor in vivo beeldvorming 19,20,21,22,23 . Eerdere protocollen hebben ofwel lps rechtstreeks geïnjecteerd in de dooier en achterhersenentrik van larvale zebravissen zonder mechanistische beoordeling, of eenvoudig LPS toegevoegd aan viswater (kweekmedium) om een dodelijke systemische immuunrespons te induceren 24,25,26,27. Hierin hebben we een protocol ontwikkeld voor micro-injectie van LPS in de hersenventrikels, om een aangeboren immuunrespons of neurotoxiciteit te activeren in de 5 dagen na de bevruchting (dpf) zebravislarven. Deze respons wordt aangetoond door neuronaal celverlies, bewegingsvermogenstekort, verhoogde afgifte van stikstofmonoxide, activering van inflammatoire genexpressie en rekrutering van neutrofielen in de hersenen van zebravissen 24 uur na injectie.
Een toenemende hoeveelheid epidemiologische en experimentele gegevens impliceert chronische bacteriële en virale infecties als mogelijke risicofactoren voor neurodegeneratieve ziekten36. De infectie veroorzaakt de activering van ontstekingsprocessen en immuunresponsen van de gastheer37. Zelfs als de reactie fungeert als een verdedigingsmechanisme, is overactiveerde ontsteking schadelijk voor neurogenese en staat de ontstekingsomgeving de overleving van pasgeboren neuronen …
