Het protocol toont aan dat door microtransplantatie van synaptische membranen in Xenopus laevis-eicellen uit te voeren, het mogelijk is om consistente en betrouwbare reacties van α-amino-3-hydroxy-5-methyl-4-isoxazolpropionzuur en γ-aminoboterzuurreceptoren vast te leggen.
Exciterende en remmende ionotrope receptoren zijn de belangrijkste poorten van ionenfluxen die de activiteit van synapsen bepalen tijdens fysiologische neuronale communicatie. Daarom zijn veranderingen in hun overvloed, functie en relaties met andere synaptische elementen waargenomen als een belangrijke correlatie van veranderingen in de hersenfunctie en cognitieve stoornissen bij neurodegeneratieve ziekten en psychische stoornissen. Begrijpen hoe de functie van exciterende en remmende synaptische receptoren wordt veranderd door ziekte is van cruciaal belang voor de ontwikkeling van effectieve therapieën. Om ziekterelevante informatie te verkrijgen, is het belangrijk om de elektrische activiteit van neurotransmitterreceptoren vast te leggen die functioneel blijven in het zieke menselijke brein. Tot nu toe is dit de dichtstbijzijnde benadering om pathologische veranderingen in de functie van receptoren te beoordelen. In dit werk wordt een methodologie gepresenteerd om microtransplantatie van synaptische membranen uit te voeren, die bestaat uit het reactiveren van synaptische membranen uit snap bevroren menselijk hersenweefsel met menselijke receptoren, door de injectie en posterieure fusie in het membraan van Xenopus laevis-eicellen . Het protocol biedt ook de methodologische strategie om consistente en betrouwbare reacties van α-amino-3-hydroxy-5-methyl-4-isoxazolepropionzuur (AMPA) en γ-aminoboterzuur (GABA) receptoren te verkrijgen, evenals nieuwe gedetailleerde methoden die worden gebruikt voor normalisatie en rigoureuze gegevensanalyse.
Neurodegeneratieve aandoeningen treffen een groot percentage van de bevolking. Hoewel hun verwoestende gevolgen bekend zijn, is het verband tussen de functionele veranderingen van neurotransmitterreceptoren, die van cruciaal belang zijn voor de hersenfunctie, en hun symptomatologie nog steeds slecht begrepen. Interindividuele variabiliteit, chronische aard van de ziekte en sluipenderwijs begin van symptomen zijn slechts enkele van de redenen die het begrip van de vele hersenaandoeningen hebben vertraagd waarbij chemische onevenwichtigheden goed gedocumenteerd zijn 1,2. Diermodellen hebben waardevolle informatie gegenereerd en onze kennis uitgebreid over de mechanismen die ten grondslag liggen aan fysiologie en pathofysiologie in evolutionair geconserveerde systemen; verschillende interspecies verschillen tussen knaagdieren en mensen verhinderen echter de directe extrapolatie van receptorfunctie van diermodellen naar het menselijk brein3. Zo werden de eerste inspanningen om inheemse menselijke receptoren te bestuderen ontwikkeld door het laboratorium van Ricardo Miledi met behulp van chirurgisch verwijderd weefsel en bevroren monsters. Deze eerste experimenten gebruikten hele membranen die neuronale synaptische en extra synaptische receptoren omvatten, evenals niet-neuronale neurotransmitterreceptoren, en hoewel ze belangrijke informatie over zieke toestanden bieden, is er bezorgdheid dat de mix van receptoren de interpretatie van gegevens bemoeilijkt 4,5,6,7. Belangrijk is dat synapsen het belangrijkste doelwit zijn bij veel neurodegeneratieve aandoeningen 8,9; daarom zijn testen om de functionele eigenschappen van aangetaste synapsen te testen van fundamenteel belang om informatie te verkrijgen over ziekterelevante veranderingen die de synaptische communicatie beïnvloeden. Hier wordt een wijziging van de oorspronkelijke methode beschreven: microtransplantatie van synaptische membranen (MSM), die zich richt op de fysiologische karakterisering van verrijkte synaptische eiwitpreparaten en met succes is toegepast om ratten en menselijke synaptosomen te bestuderen 10,11,12,13,14,15 . Met deze methodologie is het mogelijk om synaptische receptoren te transplanteren die ooit in het menselijk brein werkten, ingebed in hun eigen inheemse lipiden en met hun eigen cohort van geassocieerde eiwitten. Omdat MSM-gegevens kwantitatief zijn, is het bovendien mogelijk om deze gegevens te gebruiken om te integreren met grote proteomische of sequencinggegevenssets10.
Het is belangrijk op te merken dat veel farmacologische en biofysische analyses van synaptische receptoren worden uitgevoerd op recombinante eiwitten 16,17. Hoewel deze benadering beter inzicht biedt in de structuur-functierelaties van receptoren, kan het geen informatie geven over complexe multimere receptorcomplexen in neuronen en hun veranderingen in ziekte. Daarom moet een combinatie van inheemse en recombinante eiwitten een uitgebreidere analyse van synaptische receptoren opleveren.
Er zijn veel methoden om synaptosomen 10,11,12,13,14,15 te bereiden die kunnen worden aangepast aan de vereisten van een laboratorium. Het protocol begint met de veronderstelling dat synaptosomaal verrijkte preparaten werden geïsoleerd en klaar zijn om te worden verwerkt voor microtransplantatie-experimenten. In het laboratorium wordt de Syn-Per-methode gebruikt volgens de instructies van de fabrikant. Dit wordt gedaan vanwege de hoge reproduceerbaarheid in elektrofysiologische experimenten10,11. Er is ook overvloedige literatuur die uitlegt hoe Xenopus-eicellen 18,19 kunnen worden geïsoleerd, die ook klaar voor injectie kunnen worden gekocht20.
Analyse van inheemse eiwitcomplexen uit menselijke hersenen is nodig om homeostatische en pathologische processen bij hersenaandoeningen te begrijpen en therapeutische strategieën te ontwikkelen om ziekten te voorkomen of te behandelen.
