Hier stellen wir ein reproduzierbares In-vitro-Elektroporationsprotokoll für die genetische Manipulation von primären Kleinhirn-Granulazellvorläufern (GCPs) vor, das kostengünstig, effizient und lebensfähig ist. Darüber hinaus demonstriert dieses Protokoll auch eine einfache Methode für die molekulare Untersuchung primärer Cilium-abhängiger Hedgehog-Signalwege in primären GCP-Zellen.
