Synaptic Vesikel (SV) fietsen is het mechanisme van de kern van de intercellulaire communicatie in neuronale synapsen. FM kleurstof opname- en release zijn het belangrijkste middel van kwantitatief onderzoek SV endo- en exocytose. Hier, vergelijken wij alle stimulatie methoden om te rijden FM1-43 fietsen in de Drosophila motorische eindplaat (NMJ) model synaps.
FM kleurstoffen worden gebruikt voor het bestuderen van de synaptische Vesikel (SV) cyclus. Deze amphipathic sondes hebben een hydrofiele kop en hydrofobe staart, waardoor ze wateroplosbaar met de mogelijkheid om omkeerbaar betreden en verlaten van membraan lipide bilayers. Deze styryl-kleurstoffen zijn relatief niet-belichting fluorescerende in waterig medium, maar de invoeging in het buitenste van de bijsluiter van het plasma-membraan veroorzaakt een > 40 X verhoging van fluorescentie. In neuronale synapsen, zijn FM kleurstoffen geïnternaliseerd tijdens SV endocytose, verhandelde zowel binnen als tussen SV zwembaden en vrijgegeven met SV exocytose, bieden een krachtig hulpmiddel om te visualiseren presynaptische stadia van transmissie. Een primaire genetische model van glutamaterge synapse ontwikkeling en functie is de Drosophila motorische eindplaat (NMJ), waar FM kleurstof imaging is gebruikt uitgebreid te kwantificeren SV dynamiek in een brede waaier van mutant voorwaarden. De NMJ synaptic terminal is gemakkelijk bereikbaar, met een prachtig scala aan grote synaptic los ideaal voor beeldtoepassingen. Hier, wij vergelijken en het contrast van de drie manieren om te stimuleren de Drosophila NMJ rijden activiteit-afhankelijke FM1-43 kleurstof opname/release: 1) bad toepassing van hoge [K+] 2) zuignap elektrode motorische zenuw te depolarize neuromusculaire weefsels stimulatie om te depolarize van de presynaptische zenuw terminal, en 3) gericht transgene expressie van channelrhodopsin varianten voor licht-gestimuleerd, ruimtelijke controle van depolarisatie. Elk van deze methoden heeft voor- en nadelen voor de studie van genetische mutatie gevolgen op de SV-cyclus op de Drosophila NMJ. We zullen bespreken deze voor- en nadelen bij de selectie van de stimulatie aanpak, samen met de methoden die specifiek zijn voor elke strategie. Naast fluorescerende imaging kunnen FM kleurstoffen photoconverted aan elektron-dichte signalen gevisualiseerd met behulp van transmissie-elektronenmicroscopie (TEM) om te studeren van SV cyclus mechanismen op ultrastructureel niveau. Wij bieden de vergelijkingen van confocale en elektronenmicroscopie beeldvorming van de verschillende methoden van Drosophila NMJ stimulatie, om te helpen guide de selectie van toekomstige experimentele paradigma’s.
Het prachtig gekenmerkt Drosophila larvale motorische eindplaat (NMJ) glutamaterge synapse model is gebruikt synapse formatie en functie met een grote spectrum van genetische verstoringen1te gaan studeren. De motor neuron terminal bestaat uit meerdere takken van de axon, elk met vele uitgebreide synaptic los. Deze ruim varicosities (maximaal 5 µm in diameter) bevat alle van de transmissie-machine, met inbegrip van uniforme glutamaterge synaptische vesikels (SVs; ~ 40 nm diameter) in cytosolische reserve en gemakkelijk-vrij te geven zwembaden2. Deze blaasjes op het basisstation op de presynaptische plasmamembraan fusion site actieve zones (AZs), waar exocytose de glutamaat neurotransmitter release voor trans bemiddelt-synaptic communicatie. Vervolgens worden de SVs opgehaald uit het plasma-membraan via kus-en-run-recycling- of clathrin-gemedieerde endocytose (CME) voor herhaalde exo/endocytose cycli. De Drosophila NMJ is gemakkelijk toegankelijk en geschikt voor zowel isoleren en karakteriseren van SV cyclus mutanten. Met behulp van voorwaartse genetische schermen, hebben nieuwe mutaties geleid tot de identificatie van nieuwe genen kritisch voor de SV cyclus3. Bovendien, omgekeerde genetische benaderingen, beginnend met de reeds bekende genen hebben geleid tot de opheldering van nieuwe mechanismen van de cyclus van de SV door de zorgvuldige beschrijving van mutant fenotypen4fietsen. De Drosophila NMJ is bijna ideaal als een experimentele synaptic voorbereiding voor het ontleden van SV endocytose exocytose HORLOGEMECHANISMES via methoden om optisch track vesikel fietsen tijdens transmissie.
Een waaier van fluorescente markeringen visuele tracking van blaasjes tijdens het fietsen van dynamiek, maar de meest veelzijdige zijn FM kleurstof analogen die is voor het eerst gesynthetiseerd door Mao, F., et al.. 5. structureel, FM kleurstoffen bevatten een hydrofiele kop en een lipofiele staart verbonden via een aromatische ring, met een centrale regio spectrale eigenschappen toekennen. Deze styryl kleurstoffen partitioneren omkeerbaar in membranen, niet ‘flip-flop’ tussen membraan folders en dus nooit zijn vrij in het cytosol en zijn veel meer tl in membranen dan water5. Omkeerbare inbrengen in een lipide dubbelgelaagde veroorzaakt een verhoging van de 40-fold in fluorescentie6. Bij neuronale synapsen bestaan klassieke FM kleurstof labeling experimenten uit Baden van de synaptische voorbereiding met de kleurstof tijdens depolarizing stimulatie te laden kleurstof via SV endocytose. Externe kleurstof wordt dan gewassen weg en de SV-cyclus wordt gearresteerd in een calcium-vrije ringer-oplossing om het imago van de geladen synapsen7. Een tweede ronde van stimulatie in een bad met kleurstof-vrije triggers FM release via exocytose, een proces dat kan worden gevolgd door het meten van de daling van de intensiteit van de fluorescentie. SV populaties van een enkele blaasje tot zwembaden met honderden blaasjes kunnen kwantitatief gecontroleerde6,7. FM kleurstoffen zijn gebruikt om te ontleden activiteit-afhankelijke mobilisatie van functioneel verschillende SV zwembaden, en te vergelijken kus-en-run vs. CME fietsen8,9. De methode is aangepast om afzonderlijk assay opgeroepen, spontane en miniatuur synaptic cyclus activiteiten (met uiterst gevoelige apparatuur op te sporen van zeer kleine fluorescentie wijzigingen en verminderen photobleaching)10. Testen kunnen worden uitgebreid tot de ultrastructureel niveau door photoconverting de fluorescerende FM-signaal in een elektron-dichte label voor Transmissie Electronenmicroscopie11,12,13,14 .
Historisch, de synaptische preparaten zwemmen in een hoog gehalte aan kalium (hierna te noemen “hoge [K+]”) is de methode van keuze voor depolarizing stimulatie om te induceren SV fietsen; de kikker cholinerge NMJ5, variërend tot gekweekt knaagdier hersenen hippocampal neuronen15, de Drosophila glutamaterge NMJ model16,17. Deze hoge [K+] aanpak is eenvoudig, vereist geen gespecialiseerde apparatuur, en daarom de meeste labs toegankelijk is, maar heeft beperkingen voor zowel de toepassing en de interpretatie van de gegevens. Een veel meer fysiologisch geschikte methode is het gebruik van zuiging elektrode elektrische stimulatie van de nervus4,5,12. Deze aanpak drijft actiepotentiaal vermeerdering voor directe stimulatie van de presynaptische zenuw terminal, en resultaten kunnen direct worden vergeleken met elektrofysiologische testen van transmissie functie13,14, 15, maar vereist gespecialiseerde apparatuur en is technisch veel moeilijker. Met de komst van optogenetics heeft het gebruik van neuronale stimulatie van de channelrhodopsin extra voordelen, met inbegrip van strakke Spatio controle van kanaal expressie met behulp van de binaire Gal4/UAS systeem20. Deze aanpak is technisch veel gemakkelijker dan zuig elektrode stimulatie en vereist niets meer dan een zeer goedkope LED lichtbron. Hier, gebruiken we beeldvorming van FM1-43 (N-(3-triethylammoniumpropyl)-4-(4-(dibutylamino)styryl) pyridinium dibromide) tot en met zowel vergelijken en het contrast van deze drie methoden van de verschillende stimulatie bij de Drosophila NMJ: eenvoudige hoge [K+ ], uitdagende channelrhodopsin van elektrische en nieuwe benaderingen.
Hoge [K+] zoute depolarizing stimulatie is veruit de eenvoudigste van de drie opties voor activiteit-afhankelijke FM kleurstof fietsen, maar waarschijnlijk de minste fysiologische29. Deze eenvoudige methode depolarizes elke toegankelijk cel in het hele dier, en dus kan geen gerichte studies. Het mogelijk toe te passen lokaal hoog [K+] zoutoplossing met een micropipet, maar dit zal nog steeds depolarize pre/postsynaptisch cellen en waarschijnlijk synapse-geassocieerde glia<sup…
