We here present an optimized, detailed protocol for double immunostaining in formalin-fixed, paraffin-embedded rat central nervous system (CNS) and peripheral lymph node (LN) tissue sections.
Immunohistochimie (IHC) permet une visualisation très spécifique, fiable et attrayante protéine. Des performances correctes et l'interprétation d'un étiquetage multicolore à base d'IHC est difficile, surtout lorsqu'il est utilisé pour évaluer les interrelations entre les protéines cibles dans le tissu avec une teneur élevée en matières grasses telles que le système nerveux central (SNC).
Notre protocole représente un perfectionnement de la technique d'immuno-marquage norme particulièrement ajustée pour la détection des deux protéines structurales et solubles dans le SNC de rat et de ganglions lymphatiques périphériques (LN) affecté par la neuro-inflammation. Néanmoins, avec ou sans autres modifications, notre protocole pourrait probablement être utilisé pour la détection d'autres cibles de protéines apparentées, même dans d'autres organes et des espèces que présenté ici.
Malgré l'utilisation de pointe à haut débit les analyses effectuées sur le méthylome, transcriptome ou même au niveau du protéome, immunomarquage reste l'étalon or pour la détection de protéines directement dans l'échantillon de tissu, culture cellulaire ou un frottis cellulaire. En révélant le motif de localisation / distribution, immunohistochimie (IHC) peut évaluer les rapports relatifs et interrelations topographiques des protéines cibles, et même indiquer leurs activités biologiques. Par conséquent, IHC est largement utilisé à des fins cliniques et de recherche, par exemple, pour le diagnostic, les évaluations de traitement, l' étude des mécanismes de la maladie, des altérations fonctionnelles et phénotypiques dans des modèles animaux, etc.
Comprenant essentiellement l' histologie, la pathologie, la biochimie et l' immunologie, IHC a considérablement progressé depuis 1941, lorsque les anticorps marqués par fluorescence ont été utilisés pour la première fois pour identifier des antigènes pneumococciques dans le tissu infecté 1. Visualization des produits et des composants par IHC cellulaires est basée sur la liaison des anticorps (Abs) à leur antigène spécifique (Ag). En plus d' utiliser des anticorps fluorophores marqués, les réactions immunitaires peuvent également être visualisés à l'aide d' enzymes comme la peroxydase ou la phosphatase alcaline 2,3 4. En outre, des anticorps marqués de l' or colloïdal 5 sont utilisés pour détecter l' interaction antigène-anticorps spécifique à la fois par la lumière et par microscopie électronique, tandis que des marqueurs radioactifs sont visualisées par autoradiographie.
Le immunoréaction Ag-Ab peut être détectée par des méthodes directes et indirectes. La méthode directe est essentiellement plus rapide et plus simple, car il utilise directement marqué Abs primaire 6. Cependant, en raison du manque de sensibilité significative, les méthodes indirectes sont préférées aux directs. Procédures de détection indirecte en deux étapes nécessitent Abs primaire non marqués, comme le premier, et étiquetés Abs secondaires dirigés contre le Abs primaire, la seconde couche 7.l'amplification du signal peut être obtenue en associant en outre, une enzyme couplée tertiaire Ab (trois étapes méthode indirecte) qui se fixe à l'Ac secondaire. des procédés de détection indirecte couramment utilisés sont la biotine et l'avidine-peroxydase de antiperoxydase (PAP). Notamment, la phosphatase alcaline (AP) méthodes semblent être encore plus sensible que les méthodes d'immunoperoxydase 4. méthode de complexe avidine-biotine (ABC) utilise biotinylé Ab secondaire en combinaison avec soit étiqueté complexe avidine-biotine (LAB), ou étiqueté streptavidine-biotine complexe (SLAB). La sensibilité de détection peut être encore augmentée en y associant l' avidine marquée à la peroxydase ou la phosphatase alcaline 8. D' autres méthodes de détection utilisées sont l' étiquetage polymère, tyramine amplification et immuno-cercle roulant 9. En particulier, les différentes méthodes de détection peuvent être combinés pour la détection multiple Ag dans le même tissuéchantillon, qui a été signalé pour la première fois en 1978 4. à double immunocoloration simultanée présentée ici a été réalisée dans des coupes de tissus fixés au formol, rat de paraffine et du SNC LN en utilisant des anticorps secondaires peroxydase lié et AP-conjugués, respectivement. Les signaux ont été visualisées en utilisant 3,3'-diaminobencidine (DAB) chromogène et le Fast Blue (FB) APAAP complexe, respectivement.
les procédures IHC standard nécessitent souvent des ajustements spécifiques pour obtenir un résultat optimal, ce qui implique généralement une grande expérience, mais aussi l'approche "essais et erreurs".
