Dit protocol demonstreert methodologieën voor een muismodel van open-schedel traumatisch hersenletsel en transplantatie van gekweekte menselijke geïnduceerde pluripotente stamcel-afgeleide cellen in de wond site. Gedrags-en histologische tests van uitkomsten van deze procedures worden ook in het kort beschreven.
Traumatisch hersenletsel (TBI) is een leidende oorzaak van morbiditeit en sterfte wereldwijd. Ziekte pathologie als gevolg van TBI vordert van de primaire mechanische belediging tot secundaire letsel processen, met inbegrip van apoptosis en ontsteking. Animal Modeling is waardevol geweest in de zoektocht naar het ontrafelen van letsel mechanismen en het evalueren van potentiële neuroprotectieve therapieën. Dit protocol beschrijft het gecontroleerde corticale impact (CCI) model van focal, open-Head TBI. Specifiek worden parameters voor het produceren van een milde eenzijdige corticale verwonding beschreven. De gedrags gevolgen van CCI worden geanalyseerd met behulp van de kleefband verwijderings test van bilaterale sensorimotorische integratie. Met betrekking tot experimentele therapie voor TBI pathologie, dit protocol illustreert ook een proces voor het verplanten van gekweekte cellen in de hersenen. Neurale celculturen afgeleid van door de mens geïnduceerde pluripotente stamcellen (hiPSCs) werden gekozen voor hun potentieel om superieure functionele restauratie bij humane TBI-patiënten te vertonen. Chronische overleving van hiPSCs in de gastheer muis hersenweefsel wordt gedetecteerd met behulp van een gemodificeerde DAB immunohistochemisch proces.
Traumatisch hersenletsel (TBI) is een algemene term voor de verworven verwonding aan de hersenen als gevolg van ofwel indirecte mechanische krachten (rotatie versnelling/vertraging of contra coup) van slagen naar het hoofd of directe schade van objecten of Blast golven. Er wordt geschat dat TBI de oorzaak is van ruwweg 9% van de wereldwijde sterfgevallen en in een geschatte 50.000.000 jaar1,2wordt waargenomen. Een 2017 rapport van de centra voor ziektebestrijding en preventie geschat dat in 2013, waren er in totaal 2.800.000 ziekenhuis bezoeken en sterfgevallen als gevolg van TBI in de Verenigde Staten3. Veel mildere TBIs gaan elk jaar ongerapporteerd. Ernstige TBI kan leiden tot levenslange verslechtering van de cognitie, motorische functie, en de algehele kwaliteit van leven. De gevolgen van milde TBI, vooral repetitieve sportgerelateerde TBI, zijn pas onlangs gewaardeerd voor hun verraderlijke gezondheidseffecten4,5.
Preklinische modellering is een essentieel onderdeel van het ontwikkelen van nieuwe mechanistische inzichten en mogelijke herstellende therapie voor TBI. Het gecontroleerde corticale impact (CCI) model van TBI is een open-Head model van mechanische contusie letsel aan de cortex. De impact parameters kunnen worden aangepast om CCI letsels te produceren die variëren van mild tot ernstig6. CCI-verwondingen zijn eerder brand in plaats van diffuus, zoals te zien is bij andere gesloten hoofdmodellen van TBI. CCI kan worden uitgevoerd om een eenzijdige verwonding te induceren, zodat de contralaterale cortex kan dienen als een interne comparator. Dit protocol toont de kenmerken van een milde CCI aan een deel van de cortex dat primaire somatosensorische en motorische regio’s omvat. Dit corticale gebied werd gekozen voor zijn betrokkenheid bij sensorimotorische gedragingen waarvoor talrijke gedrags tests letsel-geïnduceerde tekorten7kunnen detecteren. Gedrags verbeteringen als gevolg van therapeutische interventies voor TBI kunnen ook worden gedetecteerd.
Een kenmerk van TBI is wijdverspreide neurale disfunctie in de benadeelde regio. Gewonde neuronen ondergaan celdood, en neuronale netwerkconnectiviteit wordt verstoord8,9. TBI verstoort de werving van endogene stamcellen, wat leidt tot verder stroomafwaarts gedrag tekorten10,11. Transplantatie van neurale stamcellen en stamcel-afgeleide cellen is onderzocht als een mogelijkheid om te herstellen van de functie in de gewonde hersenen. Naast het potentieel om beschadigde neurale circuits te herstellen, kunnen getransplanteerde cellen paracrine-effecten uitoefenen die neuronale overleving en functioneel herstel van TBI12bevorderen. Een verscheidenheid van celtypen zijn preclinically getransplanteerd om hun herstellende potentieel te evalueren in modellen van neurologische aandoeningen13,14,15. De recente popularisatie van geïnduceerde pluripotente stamceltechnologie16 heeft de ontwikkeling van talrijke menselijke stamcellijnen voor experimenteel gebruik vergemakkelijkt. Preklinisch onderzoek met hiPSC-afgeleide cellen is een belangrijke eerste stap om de potentiële therapeutische werkzaamheid van een bepaalde cellijn tegen menselijke ziektes te karakteriseren. Dit laboratorium heeft protocollen ontwikkeld om hiPSCs te differentiëren naar neurale fenotypes17 in het nastreven van transplanteer bare cellen om herstel van traumatisch hersenletsel te helpen.
Experimenten in dit protocol gebruiken een unilaterale CCI om TBI te induceren naar de linker somatosensorische en motorische cortex van volwassen muizen. Een milde CCI-blessure resulteert in een langdurig functioneel tekort in de rechter voorpoot dat wordt gebruikt om de effecten van de door hiPSC afgeleide neurale cel-engraftment op functioneel herstel bij te houden. Forepaw sensomotorische testen in dit protocol werd aangepast aan de methodologie van Bouet en collega’s18 en werd eerder gedemonstreerd door Fleming en collega’s19. Dit protocol schetst een complete workflow voor het uitvoeren van een experimenteel hersenletsel, therapeutische transplantatie van heup cellen en gedrags-en histologische analyse van experimentele uitkomst maatregelen.
Mild CCI als modelsysteem voor het testen van experimentele regeneratieve therapie
Het CCI-model is een waardevol hulpmiddel voor het onderzoeken van mechanismen van weefsel disfunctie na mechanisch letsel aan de cortex. De instelbaarheid van de blessure parameters is een aantrekkelijk kenmerk van dit model.
