Organic dye molecules and oleic acid coated upconverting nanoparticles are not water-soluble. This protocol describes a ‘plug and play’ method that enables the transfer of organic dye molecules and upconverting particles from their initial hydrophobic solvent to water.
In this protocol, we first describe a procedure to synthesize lanthanide doped upconverting nanoparticles (UCNPs). We then demonstrate how to generate amphiphilic polymers in situ, and describe a protocol to encapsulate the prepared UCNPs and different organic dye molecules (porphyrins and diarylethenes) using polymer shells to form stable water-dispersible nanoassemblies. The nanoassembly samples containing both the UCNPs and the diarylethene organic dyes have interesting photochemical and photophysical properties. Upon 365 nm UV irradiation, the diarylethene group undergoes a visual color change. When the samples are irradiated with visible light of another specific wavelength, the color fades and the samples return to the initial colorless state. The samples also emit visible light from the UCNPs upon irradiation with 980 nm near-infrared light. The emission intensity of the samples can be tuned through alternate irradiation with UV and visible light. Modulation of fluorescence can be performed for many cycles without observable degradation of the samples. This versatile encapsulation procedure allows for the transfer of hydrophobic molecules and nanoparticles from an organic solvent to an aqueous medium. The polymer helps to maintain a lipid-like microenvironment for the organic molecules to aid in preservation of their photochemical behavior in water. Thus this method is ideal to prepare water-dispersible photoresponsive systems. The use of near-infrared light to activate upconverting nanoparticles allows for lower energy light to be used to activate photoreactions instead of more harmful ultraviolet light.
Vandaag is er nog steeds een dringende behoefte aan nieuwe vormen van bio-imaging agenten te ontwikkelen. Een andere benadering voor fluorescentie moduleren koppelen licht aansprekend chromoforen met fluorescerende probes. 15,16 omschakelen het licht aansprekend chromofoor tussen twee isomeren waarin slechts één van de isomeren kan fungeren als een efficiënte energieoverdracht acceptor, maakt controle uitdoving van de fluorescentie van the sonde door middel van Förster Resonance Energy Transfer (FRET) en andere mechanismen. Het resultaat is de creatie van een emitterende staat en een uitgeblust staat die kan worden afgewisseld door blootstelling van de fotoresponsieve chromofoor op verschillende golflengten van het licht.
Fotoresponsieve diarylethene chromoforen kunnen reversibel worden omgeschakeld tussen een kleurloze ring-open isomeer en een gekleurde ring-gesloten isomeer na bestraling met UV en zichtbaar licht. 17-19 De thermische stabiliteit van de twee isomeren en afstembare absorptiespectra van de ringgesloten isomeer make diarylethenen zeer goede kandidaten als controleerbaar FRET acceptoren. 20-23 Lanthanide-gedoteerde NaYF 4 upconverting nanodeeltjes zijn nuttig voor bio-imaging. 24 Deze nanodeeltjes absorberen nabij-infrarood licht en licht uitzenden in verschillende regio's van het zichtbare spectrum. Voorbeelden van fluorescentie modulatie combineert licht aansprekend diarylethene chromoforen en nanodeeltjes zijn previously gerapporteerd door de groep. 25-27 De in elk voorbeeld beschreven systemen vereist een extra synthetische wijziging van de diarylethenen hechten aan het oppervlak van de nanodeeltjes, die de ontwikkeling van diverse systemen bemoeilijkt.
Hierin tonen we een eenvoudige 'plug-and-play' methode om waterdispergeerbare organische kleurstofmoleculen en fotoresponsieve upconverting nanodeeltjes door middel van een zelf-assemblage strategie voor te bereiden. De keuze van polymeren; poly (styreen-alt-maleïnezuuranhydride) en polyetheramine 2070 bieden zowel een hydrofobe als hydrofiele omgeving. De hydrofobe gedeelten van het polymeer helpen om de normaal water oplosbare organische moleculen en omhoog converteren van nanodeeltjes samenhouden, terwijl de hydrofiele regio van het polymeer is essentieel voor het handhaven van de oplosbaarheid in water. We zullen eerst demonstreren de synthese van de up-converting nanodeeltjes door de thermische kiemvorming methode. Dan zullen we bewijzen how de organische moleculen en omhoog converteren van nanodeeltjes zijn ingekapseld in hydrofobe gebieden van de polymeermantel en stabiel in waterige media blijven door eenvoudigweg samen roeren van een oplossing van de upconverting nanodeeltjes, polymeren en andere organische kleurstofmoleculen, gevolgd door een geschikte opwerkprocedure. We tonen ook aan hoe fluorescentie-emissie van de samenstellen met behulp van externe bestraling met licht te moduleren. We anticiperen op de omvang van het gebruik van deze 'plug-and-play' methode om waterdispergeerbaar nanoassemblies zal blijven uitbreiden.
