This protocol describes simultaneous measurement of electroretinogram and visual evoked potentials in anesthetized rats.
The electroretinogram (ERG) and visual evoked potential (VEP) are commonly used to assess the integrity of the visual pathway. The ERG measures the electrical responses of the retina to light stimulation, while the VEP measures the corresponding functional integrity of the visual pathways from the retina to the primary visual cortex following the same light event. The ERG waveform can be broken down into components that reflect responses from different retinal neuronal and glial cell classes. The early components of the VEP waveform represent the integrity of the optic nerve and higher cortical centers. These recordings can be conducted in isolation or together, depending on the application. The methodology described in this paper allows simultaneous assessment of retinal and cortical visual evoked electrophysiology from both eyes and both hemispheres. This is a useful way to more comprehensively assess retinal function and the upstream effects that changes in retinal function can have on visual evoked cortical function.
Måling af elektroretinogrammet (ERG) og visuelt fremkaldt potentiale (VEP) give nyttige kvantitative vurderinger af integriteten af den visuelle pathway. ERG måler de elektriske reaktioner i nethinden for lys stimulation, mens VEP måler den tilsvarende funktionelle integritet af de visuelle veje fra nethinden til den primære visuelle cortex efter samme lys begivenhed. Dette håndskrift beskriver en protokol til registrering og analyse af ERG og VEP reaktioner i et almindeligt anvendt laboratorium model, rotten.
ERG tilvejebringer et indeks for den funktionelle integritet en række centrale retinale celle klasser ved at kvantificere retinas brutto elektriske reaktion på et lysglimt. Dette håndskrift fokuserer på metode til måling af ERG og analysere dens funktioner til at returnere information om nogle af de store celle klasser i nethinden, nemlig fotoreceptorer (den PIII komponent), bipolære celler (PIO komponent) og retinale ganglieceller (den positive scotopic tærskel respons eller pSTR).

VEP, når den anvendes sammen med ERG, kan tilvejebringe en mere fuldstændig vurdering af det visuelle system (dvs. retina / visuelle pathway).ERG og VEP optagelser kan udføres isoleret eller i kombination, afhængig af ansøgkation. Den i dette dokument metodik muliggør samtidig evaluering af retinal og kortikal visuelt fremkaldt elektrofysiologi fra begge øjne og begge halvkugler i bedøvede rotter. ERG og VEP er objektive mål for visuel funktion fra nethinden og cortex, hhv. Fordelen ved samtidig optagelse er, at en mere omfattende udsigt over hele visuelle pathway, der ydes.
