Analyzing the physiological properties of olfactory sensory neurons still faces technical limitations. Here we record them through perforated patch-clamp in an intact preparation of the olfactory epithelium in gene-targeted mice. This technique allows the characterization of membrane properties and responses to specific ligands of neurons expressing defined olfactory receptors.
Het analyseren van de fysiologische reacties van olfactorische sensorische neuronen (OSN) wanneer gestimuleerd met specifieke liganden is van cruciaal belang om de basis van olfactorische-gedreven gedrag en hun modulatie begrijpen. Deze coderende eigenschappen hangen sterk af van de initiële interactie tussen geurmoleculen en de olfactorische receptor (OR) uitgedrukt in de ASN. De identiteit, specificiteit en ligand spectrum van de uitgedrukt of zijn kritisch. De kans op het ligand van de OR uitgesproken vinden in een OSN binnen het epitheel willekeurig gekozen is zeer laag. Om deze uitdaging aan te pakken, dit protocol maakt gebruik van genetisch gelabelde muizen die het fluorescente eiwit GFP onder de controle van de promotor van gedefinieerde ultraperifere regio's. OSNs wordt in een strakke en georganiseerde epitheel langs de neusholte met naburige cellen hun rijping en functie beïnvloeden. Hier wordt een methode beschrijven we een intacte reukepitheel te isoleren en op te nemen door middel van patch-clamp opnames van de eigenschappen van OSNs expressing gedefinieerd geurstof receptoren. Het protocol kan men OSN membraaneigenschappen karakteriseren terwijl de invloed van het naburige weefsel. Analyse van de patch-clamp resultaten levert een exacte kwantificering van ligand / of interacties, transductieroutes en farmacologie, OSNs 'codering eigenschappen en hun modulatie op het membraan niveau.
Olfactorische sensorische neuronen (OSN) vormen de eerste stap van olfactorische waarneming. Gelegen in de olfactorische epithelium langs de neusholte bij knaagdieren, zij de chemische karakterisering van geurstoffen in maatregelen potentialen die via de axon naar de hersenen. Om een beter inzicht olfactorische coderingsmechanismen, moet de transductie en membraaneigenschappen van OSNs karakteriseren. Tot voor kort waren de meeste van de technieken om de eigenschappen van zoogdier OSNs karakteriseren werden op gedissocieerd OSNs 1-4 uitgevoerd. De dissociatie proces maakt gebruik van verschillende mechanische en chemische (dat wil zeggen, enzymen) processen om de ASN's te bevrijden uit hun omgeving. Deze werkwijzen leiden tot een lage aantal beschikbare cellen voor opnames. Dit lage aantal kan nog kritischer bij GFP gemerkte cellen. Dissociatie wordt ook de lokale cel-cel interacties tussen ASN's en andere cellen van de reukepitheel die overleefde kunnen versterkenal en modulatie van eigenschappen OSNs '. Om de dissociatie procedure omzeilen, werd een intact preparaat ontwikkeld 5.
Elke OSN drukt een olfactorische receptor (OR) geselecteerd uit een groot multigenfamilie 6. Er zijn ~ 1000 UPR uitgedrukt in de belangrijkste olfactorisch epitheel bij de muis. Vanwege het grote aantal OR in wildtype dieren, de kansen om ASN expressie dezelfde record of zeer laag. Om deze beperkingen te overwinnen, gen gerichte muizen zijn verkrijgbaar waarbij alle OSNs expressie een bepaalde of gelabeld met een fluorescent proteïne 7-9. Deze gelabelde OSNs werden gebruikt om functionele analyse doen in gedissocieerde voorbereidingen 7,10,11 met de eerder genoemde nadelen. Een intacte epitheel voorbereiding 5 van genetisch gelabelde muizen omzeilt daarom deze kwesties. Het laat de controle van de activiteit van OSNs uiten precies gedefinieerde ultraperifere regio's in een omgeving die zo dicht mogelijk bij in vivo mogelijk. Trouwens, patch-clamp opnames van OSNs ook mogelijk nauwkeurige analyse van de membraan eigenschappen, transductieroute farmacologie, ligand / of interacties. Al deze onderwerpen kunnen nauwelijks worden geanalyseerd met behulp van extracellulaire opnames. We gebruikten deze techniek om de reacties van OSNs uiting van de geurstof receptoren SR1 en MOR23 12,13 bewaken. De haalbaarheid van deze techniek werd bevestigd door andere groepen MOR23 expressie OSNs 14 alsmede andere UPR expressie neuronen 15,16. Het bewaken van een bepaalde populatie OSNs kan leiden tot de analyse van de eigenschappen in ander verband zoals de ontwikkeling 14, verouderen 17, geurstof geïnduceerde plasticiteit 18, en de rol van de wijzigingen van Type de geurstof receptor in geur codering 15. Dit protocol geeft dus een krachtig instrument om het toezicht op de functionele eigenschappen van gedefinieerde OSNs op het membraan niveau.
Het vermogen van dit protocol om de eigenschappen van een gezonde OSNs correct monitoren sterk afhankelijk van de kwaliteit van de voorbereiding. Daarom zijn de dissectie stappen kritisch. Ten eerste is het essentieel om aandacht te besteden aan de kwaliteit (pH, osmolariteit), zuurstof en temperatuur (ijskoud maar niet ingevroren) van de dissectie medium. Ten tweede moet de manipulatie van het epitheel ontleden gereedschappen zo beperkt mogelijk schade te voorkomen. Tenslotte is het essentieel om een preparaat zo…
