Primaire microgliaculturen worden vaak gebruikt om nieuwe ontstekingsremmende moleculen te evalueren. Het huidige protocol beschrijft een reproduceerbare en relevante methode om microglia magnetisch te isoleren van pasgeboren pups.
Microglia, als hersenbewoner macrofagen, zijn fundamenteel voor verschillende functies, waaronder reactie op omgevingsstress en hersenhomeostase. Microglia kunnen een groot spectrum van activeringsfenotypen aannemen. Bovendien worden microglia die het pro-inflammatoire fenotype ondersteunen geassocieerd met zowel neurologische als neurodegeneratieve aandoeningen. In vitro studies worden veel gebruikt in onderzoek om potentiële therapeutische strategieën in specifieke celtypen te evalueren. In deze context is het bestuderen van microgliale activering en neuro-inflammatie in vitro met behulp van primaire microgliale culturen relevanter dan microgliale cellijnen of van stamcellen afgeleide microglia. Het gebruik van sommige primaire culturen kan echter lijden aan een gebrek aan reproduceerbaarheid. Dit protocol stelt een reproduceerbare en relevante methode voor om microglia magnetisch te isoleren van pasgeboren pups. Microgliale activering met behulp van verschillende stimuli na 4 h en 24 h door mRNA-expressiekwantificering en een Cy3-kraal fagocytische assay wordt hier gedemonstreerd. Het huidige werk zal naar verwachting een gemakkelijk reproduceerbare techniek bieden voor het isoleren van fysiologisch relevante microglia uit juveniele ontwikkelingsstadia.
Microglia zijn de macrofaagachtige cellen van het centrale zenuwstelsel die zijn afgeleid van erytropoëtische voorlopers van de dooierzak die migreren naar het neuro-epitheel tijdens de vroege embryonale ontwikkeling1. Afgezien van hun immuniteitsfuncties, spelen ze ook een belangrijke rol tijdens de neurologische ontwikkeling, met name voor synaptogenese, neuronale homeostase en myelinisatie2. Op volwassen leeftijd ontwikkelen microglia lange cellulaire processen om de omgeving continu te scannen. In het geval van homeostaserupturen zoals hersenletsel of hersenziekte, kunnen microglia hun morfologische uiterlijk veranderen om een amoeboïde vorm aan te nemen, naar het gewonde gebied te migreren, veel cytoprotectieve of cytotoxische factoren te verhogen en vrij te geven. Microglia hebben heterogene activeringstoestanden, afhankelijk van hun ontwikkelingsstadium en het type opgelopen letsel 3,4,5. In deze studie worden deze activeringstoestanden grofweg ingedeeld in drie verschillende fenotypen: pro-inflammatoire / fagocytische, ontstekingsremmende en immunoregulerende, rekening houdend met het feit dat de situatie in werkelijkheid waarschijnlijk complexer is6.
Het bestuderen van in vivo microgliale activering en screening op neuroprotectieve strategieën in vroege stadia van de ontwikkeling van de hersenen kan een uitdaging zijn vanwege (1) de kwetsbaarheid van dieren vóór het spenen en (2) het lage aantal microgliale cellen. Daarom worden in vitro studies op microglia veel gebruikt voor toxiciteit 7,8,9, neuroprotectieve strategieën 5,10,11,12,13,14 en co-culturen 15,16,17,18,19,20,21 . In vitro studies kunnen microgliale cellijnen, van stamcellen afgeleide microglia of primaire microgliacultuur gebruiken. Al deze benaderingen hebben voor- en nadelen en de keuze hangt af van de initiële biologische vraag. De voordelen van het gebruik van primaire microgliaculturen zijn de homogene genetische achtergrond, pathogeenvrije geschiedenis en controle van de tijd waarin de microglia worden gestimuleerd na de dood van het dier22.
In de loop der jaren werden verschillende methoden (flowcytometrie, schudden of magnetisch labelen) ontwikkeld voor het kweken van primaire microglia van knaagdieren, zowel pasgeborenen als volwassen 23,24,25,26,27,28,29. In het huidige werk wordt microglia-isolatie van muisneaatpups uitgevoerd met behulp van eerder beschreven magnetisch geactiveerde celsorteertechnologie met behulp van microbead-gecoate anti-muis CD11b 25,27,29. CD11b is een integrine-receptor die tot expressie komt aan het oppervlak van myeloïde cellen, waaronder microglia. Wanneer er geen ontstekingsuitdaging in de hersenen is, zijn bijna alle CD11b + -cellen microglia30. Vergeleken met andere eerder gepubliceerde methoden 23,24,25,26,27,28,29, balanceert het huidige protocol onmiddellijke ex vivo microgliale activeringsanalyses en gemeenschappelijke in vitro primaire microgliale cultuur. Microglia worden dus (1) geïsoleerd op postnatale dag (P)8 zonder myelineverwijdering, (2) gekweekt zonder serum en (3) blootgesteld aan siRNA, miRNA, farmacologische verbinding en/of ontstekingsprikkels slechts 48 uur na hersenisolatie. Elk van deze drie aspecten maakt het huidige protocol relevant en snel. Allereerst maakt het gebruik van pediatrische microglia het mogelijk om dynamische en reactieve levensvatbare cellen in cultuur te verkrijgen zonder dat een extra demyelinisatiestap nodig is die mogelijk de microgliale reactiviteit in vitro zou kunnen wijzigen. Het huidige protocol heeft als doel om zo dicht mogelijk bij de fysiologische omgeving van microglia te komen. Inderdaad, microglia komen nooit serum tegen, en dit protocol vereist ook niet het gebruik van serum. Bovendien voorkomt het blootstellen van microglia al 48 uur na het kweken dat ze hun fysiologische vermogens verliezen.
Het huidige werk presenteert een primaire microgliale celcultuur met behulp van magnetisch gesorteerde CD11b + -cellen. Naast de microgliale functionele evaluatie (RT-qPCR en fagocytische assays) werd ook de microgliale cultuurzuiverheid bepaald.
