This manuscript provides a step-by-step procedure for the derivation and maintenance of human keratinocytes from plucked hair and subsequent generation of integration-free human induced pluripotent stem cells (hiPSCs) by episomal vectors.
Recent advances in reprogramming allow us to turn somatic cells into human induced pluripotent stem cells (hiPSCs). Disease modeling using patient-specific hiPSCs allows the study of the underlying mechanism for pathogenesis, also providing a platform for the development of in vitro drug screening and gene therapy to improve treatment options. The promising potential of hiPSCs for regenerative medicine is also evident from the increasing number of publications (>7000) on iPSCs in recent years. Various cell types from distinct lineages have been successfully used for hiPSC generation, including skin fibroblasts, hematopoietic cells and epidermal keratinocytes. While skin biopsies and blood collection are routinely performed in many labs as a source of somatic cells for the generation of hiPSCs, the collection and subsequent derivation of hair keratinocytes are less commonly used. Hair-derived keratinocytes represent a non-invasive approach to obtain cell samples from patients. Here we outline a simple non-invasive method for the derivation of keratinocytes from plucked hair. We also provide instructions for maintenance of keratinocytes and subsequent reprogramming to generate integration-free hiPSC using episomal vectors.

I motsats till blodkroppar och huden fibroblast som kräver invasiva metoder för extraktion, keratinocyter representerar en lättillgänglig celltyp för hiPSC generation. Keratinocytes är keratin-rika epitelceller som bildar den yttre epidermala barriären i huden och är även hittats i naglar och hår 11. I synnerhet kan keratinocyter hittas på den yttre rotskidan (ORS) av hårsäckar, en extern cellulär skikt som täckte hårstrået tillsammans med inner rotskidan (IRS) celler (12, figur 1). Eftersom hår kollektionen är en enkel procedur som inte kräver hjälp av medicinsk personal, ger det en möjlighet för patienter att samla in och skicka sina egna hårprover till laboratorier, som i hög grad skulle underlätta insamlingen av patientprover för hiPSC generation. Epidermala keratinocyter har också en högre omprogrammering effektivitet och snabbare omprogrammering kinetik jämfört med fibroblaster, lägga till fördelarna med att använda keratinocyter som start celler för hiPSC generation 9,13. Dessutom kan hiPSCs även genereras med hjälp av andra cellpopulationer i hårsäcken,inklusive dermalpapillen cellerna placerade vid basen av hårsäcken 14,15.
Tidigare rapporter IPSC generation använder hår-härledda celler utnyttjar ofta retrovirala eller lentivirala baserade omprogrammering metoder 9,14,15. Dessa virala metoder införa oönskad genomisk integrering av utländska transgener under omprogrammering. I jämförelse, användning av episomala vektorer representerar en möjlig, icke-viral omprogrammering metod för att generera integrationsfria iPSCs 4. Vi har tidigare utvecklat en enkel, kostnadseffektiv och icke-virala metod för att på ett effektivt sätt programmera keratinocyt i hiPSCs använder episomala vektorer 13. Här ger vi ett detaljerat protokoll för alstring av keratinocythärledda hiPSCs, inklusive härledning av keratinocyter från plockade hår, expansion och underhåll av keratinocyterna och efterföljande omprogrammering för att generera hiPSCs.
Generation av patientspecifika hiPSCs erbjuder en unik metod för att studera patogenesen i sjuka celltyper in vitro, och ger också en plattform för läkemedelsscreening för att identifiera nya molekyler som kan rädda fenotyperna sjukdomstillstånd.
