Candida albicans hyphal morfogenese i mave (GI) tarmkanalen er stramt kontrolleret af forskellige miljømæssige signaler, og spiller en vigtig rolle i formidling og patogenese af denne opportunistiske svampepatogen. Men metoder til at visualisere svampehyfa i mave-tarmkanalen in vivo er udfordrende, hvilket begrænser forståelsen af miljøsignaler i at kontrollere denne morfogenese proces. Den protokol, der er beskrevet her, viser en ny ex vivo-metode til visualisering af hyphal morfosese i guthomogene ekstrakter. Ved hjælp af en ex vivo assay, denne undersøgelse viser, at cecal indhold fra antibiotika behandlede mus, men ikke fra ubehandlede kontrol mus, fremme C. albicans hyphal morfogens i tarmen indhold. Endvidere, tilføje tilbage specifikke grupper af tarm metabolitter til cecal indholdet fra antibiotika-behandlede mus differentieret regulerer hyphal morfogenese ex vivo. Samlet set repræsenterer denne protokol en ny metode til at identificere og undersøge de miljøsignaler, der styrer C. albicans hyphal morfogens i mave-tarmkanalen.
Candida albicans er en opportunistisk, polymorfe svampepatogen, der normalt commensal, men kan gennemgå en morfologisk ændring i en virulent form i stand til at forårsage livstruende infektioner i immunkompromitterede personer1,2,3,4,5,6,7,8,9,10,11,12,13. C. albicans er en førende årsag til systemiske nosokomielle infektioner, med en 40\u201260% dødelighed selv med svampedræbende behandling2,14,15. Selvom C. albicans bor i forskellige værtsnicher, herunder det kvindelige reproduktive system16,17, stammer mundhulen for raske personer18 og mave-tarmkanalen (GI)19,20, størstedelen af de systemiske infektioner stammer fra mave-tarmkanalen og desuden, kilden til systemisk infektion bekræftes ofte som mave-tarmkanalen21,22,23,24,25,26,27,28,29,30,31,32,33,34. C. albicanspatogenicitet i mave-tarmkanalen påvirkes af en lang række faktorer; en vigtig egenskab, der er nødvendig for virulens, er imidlertid overgangen fra en gærcellemorfologi til en virulent hyphalcellemorfologi35,36,37,38,39,40,41,42,43,44. C. albicans vedhæftet fil og formidling fra mave-tarmkanalen under infektion er stærkt forbundet med sin evne til at skifte fra en commensal gær til virulent hyphae, så svampene kan forårsage invasiv sygdom44,45,46,47,48,49,50,51,52,53.
En række faktorer i tarmen, herunder n-acetylglucosamin, regulere hyphal dannelse af C. albicans. Det er derfor afgørende at mindske kløften i viden om hyphal morfogens af dette svampepatogen imave-tarmkanalen 54,55,56. De seneste beviser tyderpå,at forskellige gutmetabolitter forskelligt kontrollerer hyphal morfogenese af C. albicans in vitro57,58,59,60. Men, tekniske begrænsninger præsentere spørgsmål, når de forsøger at studere C. albicans hyphae dannelse i in vivo tarm prøver, især farvning gær og hyphae celler og kvantitativ analyse af hyphal udvikling. For at forstå C. albicans hyphal morfogens i mave-tarmkanalen, en ex vivo metode blev udviklet ved hjælp af opløselige ekstrakter af homogeniseret tarmindhold fra mus til at undersøge effekten af metabolitter på svampehyphal morfosse. Udnytte tarmprøver fra mus, der er resistente og modtagelige for C. albicans GI-infektion, denne metode vil bidrage til at identificere og studere effekten af metabolitter, antibiotika og xenobiotika på svampehyphal morfogenese i mave-tarmkanalen.
