Hier stellen wir zwei Protokolle vor, die die Modellierung von intestinalen apikal-spezifischen Interaktionen ermöglichen. Organoid-abgeleitete intestinale Monoschichten und Air-Liquid-Interface (ALI)-Kulturen erleichtern die Erzeugung von gut differenzierten Epithelien, die sowohl von luminaler als auch von basolateraler Seite zugänglich sind, während polaritätsinvertierte Darmorganoide ihre apikale Seite freilegen und für Hochdurchsatz-Assays zugänglich sind.
