Wir beschreiben ein Protokoll für die Herstellung von gRNA Bibliotheken basierend auf enzymatische Verdauung von Ziel DNA. Diese Methode CORALINA (umfassende gRNA Bibliothek Generation durch kontrollierte Nuklease Aktivität) stellt eine Alternative zu teuren benutzerdefinierte Oligonukleotid-Synthese.
