Methoden voor het opwekken van grootschalige gRNA bibliotheken moet eenvoudig, efficiënt en kosteneffectief zijn. Beschrijven we een protocol voor de productie van gRNA bibliotheken gebaseerd op enzymatische spijsvertering van DNA van het doel. Deze methode, CORALINA (uitgebreide gRNA bibliotheek generatie via gecontroleerde nuclease activiteit) presenteert een alternatief voor dure aangepaste oligonucleotide synthese.
