Her præsenteres et TIRF-mikroskopibaseret in vitro-rekonstitutionsassay for samtidig at kvantificere og sammenligne dynamikken i to mikrotubulipopulationer. En metode er beskrevet til samtidig at se den kollektive aktivitet af flere mikrotubuli-associerede proteiner på tværbundne mikrotubuli bundter og enkelte mikrotubuli.
