Hier presenteren we protocollen voor de synthese van disaccharide nucleosiden door de regioselectieve O– glycosylatie van ribonucleosides via een tijdelijke bescherming van hun 2′, 3′-diol wordt met behulp van een cyclische ester van koppeling. Deze methode is van toepassing op verschillende onbeschermde nucleosiden zoals adenosine, calciumguanosine, cytosinetrifosfaat, uridine, 5-methyluridine en 5-fluorouridine te geven van de corresponderende disaccharide nucleosiden.
Disaccharide nucleosiden, die bestaan uit disaccharide en nucleobase wordt, zijn gekend als een waardevolle groep van natuurproducten met veelsoortige bioactivities. Hoewel chemische O– glycosylation een algemeen positieve strategie is te synthetiseren disaccharide nucleosiden, vereist de voorbereiding van substraten zoals glycosyl donors en acceptanten vervelend bescherming manipulaties van de groep en een zuivering op elke synthetische stap. Ondertussen verschillende onderzoeksgroepen hebben gemeld dat koppeling en borinic esters daarvan dienen als een bescherming of groep van koolhydraten derivaten om de regio – en/of Stereoselectieve acylering, alkylatie silyl en glycosylatie activeren. In dit artikel tonen we de procedure voor de regioselectieve O– glycosylatie van onbeveiligde ribonucleosides met behulp van koppeling zuur. De verestering van 2′, 3′-diol van ribonucleosides met koppeling zuur maakt de tijdelijke bescherming van diol, en volgende O– glycosylation met een glycosyl donor in het bijzijn van de p– toluenesulfenyl chloride en zilver triflaat, vergunningen de regioselectieve reactie van de 5′-hydroxylgroep veroorloven het disaccharide nucleosiden. Deze methode kan worden toegepast op verschillende nucleosiden, zoals calciumguanosine, adenosine, cytosinetrifosfaat, uridine, 5-metyluridine en 5-fluorouridine. In dit artikel en de bijbehorende video vertegenwoordigen nuttige (visuele) informatie voor de O– glycosylatie van onbeschermde nucleosiden en hun analogen voor de synthese van niet alleen disaccharide nucleosiden, maar ook een verscheidenheid aan biologisch relevante derivaten.
Disaccharide nucleosiden, die geconjugeerde van een nucleoside en een groep koolhydraten zijn met elkaar verbonden via een O-glycosidic bond, vormen een waardevolle klasse van natuurlijk voorkomende koolhydraten derivaten1,2 ,3,4,,5,,6,7. Bijvoorbeeld, zij zijn opgenomen in de biologische macromoleculen zoals tRNA (transfer RNA zuur) en poly(ADP-ribose) (ADP = adenosinedifosfaat), evenals in sommige antibacteriële middelen en andere biologisch-werkzame stoffen (b.v., adenophostins, amicetins, ezomycin)5,6,8,9,10,11,12,13, 14,,15,16,17,18,19. Vandaar, disaccharide nucleosiden en derivaten daarvan naar verwachting loodverbindingen voor ontdekking drugsonderzoek. De methoden voor de synthese van disaccharide nucleosiden worden ingedeeld in drie categorieën; enzymatische O– glycosylation20,21, chemische N– glycosylation5,9,16,22,23, 24, en chemische O– glycosylation7,9,14,16,18,19,24, 25,26,27,28,29,30,31,32,33, 34,35,,36,,37. In het bijzonder zou chemische O– glycosylation een efficiënte methode voor de Stereoselectieve synthese en grootschalige synthese van disaccharide nucleosiden. Eerder onderzoek heeft aangetoond dat de O– glycosylatie van 2′-deoxyribonucleoside 2 met de thioglycosyl donor 1, met behulp van de combinatie van p– toluenesulfenyl chloride en zilver triflaat, biedt de gewenst disaccharide nucleoside 3 (Figuur 1A; AR = aryl en PG = bescherming groep)38.
Naar aanleiding van deze resultaten besloten we te ontwikkelen van de O– glycosylatie van ribonucleosides p– toluenesulfenyl chloride/zilver triflaat promotor systeem toe te passen. Hoewel diverse voorbeelden van de O– glycosylatie van gedeeltelijk beschermde ribonucleosides aangetoonde7,9,14,16,18,geweest19 ,24,32,33,34,35,,36,,37, het gebruik van onbeschermde of tijdelijk beveiligde ribonucleosides als een glycosyl accepteerder voor O– glycosylation verwaarloosbaar is gemeld. Daarom zou de ontwikkeling van de regioselectieve O– glycosylatie van onbeschermde of tijdelijk beveiligde ribonucleosides bieden van een gunstiger synthetische methode zonder de bescherming van de manipulaties van de groep van ribonucleosides. Met het oog op de regioselectieve O– glycosylatie van ribonucleosides, we gericht op de boor-verbindingen, omdat diverse voorbeelden van regio – en/of Stereoselectieve acylering, alkylatie silyl en glycosylatie van koolhydraten derivaten bijgestaan door koppeling of borinic zuur geweest gemeld39,40,41,42,43,44,45 ,46,47,48,49,50. In dit artikel tonen we de procedure voor de synthese van disaccharide nucleosiden gebruik te maken van de regioselectieve O– glycosylatie op de 5′-hydroxylgroep van ribonucleosides via een tussenliggende koppeling ester. In de hier gepresenteerde strategie koppeling ester tussenliggende 6 zou worden geboden door de verestering van de ribonucleoside 4 met de koppeling zure 5, waarmee de regioselectieve O– glycosylatie op de 5′-hydroxylgroep met thioglycosyl donor 7 te geven van het disaccharide nucleoside 8 (Figuur 1B)51. We bestudeerde ook de interactie van een ribonucleoside en de koppeling zuur door nucleaire magnetische resonantie (NMR) spectroscopie, te observeren van de vorming van een koppeling ester. Verestering kan een koppeling ester en een glycosylatie reactie vereisen watervrije voorwaarden om te voorkomen dat de hydrolyse van de koppeling ester en de donor glycosyl. In dit artikel tonen we de typische procedures voor het verkrijgen van de watervrije voorwaarden voor succesvolle glycosylatie reacties voor onderzoekers en studenten niet alleen in de chemie, maar ook op andere onderzoeksterreinen.
Het doel van dit manuscript is om te laten zien een handige synthetische methode te bereiden disaccharide nucleosiden met behulp van onbeveiligde ribonucleosides zonder vervelende bescherming manipulaties van de groep. Wij rapporteren hierin over de regioselectieve O– glycosylations van nucleosiden via de tijdelijke 2′, 3′-diol bescherming door een cyclische koppeling ester (Figuur 1B)51.
