Environmental enrichment provides a potential protective effect against neurodegenerative disorders. Currently, however, there is no easy way of determining the efficacy of enrichment procedures. This protocol describes a simple “Puzzle Box” method for assessing an animal’s cognitive function, in order to reveal the effectiveness of environmental enrichment.
Environmental enrichment can dramatically influence the development and function of neural circuits. Further, enrichment has been shown to successfully delay the onset of symptoms in models of Huntington’s disease 1-4, suggesting environmental factors can evoke a neuroprotective effect against the progressive, cellular level damage observed in neurodegenerative disorders. The ways in which an animal can be environmentally enriched, however, can vary considerably. Further, there is no straightforward manner in which the effects of environmental enrichment can be assessed: most methods require either fairly complicated behavioral paradigms and/or postmortem anatomical/physiological analyses. This protocol describes the use of a simple and inexpensive behavioral assay, the Puzzle Box 5-7 as a robust means of determining the efficacy of increased social, sensory and motor stimulation on mice compared to cohorts raised in standard laboratory conditions. This simple problem solving task takes advantage of a rodent’s innate desire to avoid open enclosures by seeking shelter. Cognitive ability is assessed by adding increasingly complex impediments to the shelter’s entrance. The time a given subject takes to successfully remove the obstructions and enter the shelter serves as the primary metric for task performance. This method could provide a reliable means of rapidly assessing the efficacy of different enrichment protocols on cognitive function, thus paving the way for systematically determining the role specific environmental factors play in delaying the onset of neurodevelopmental and neurodegenerative disease.
Omgevingsverrijking (EE) kan worden gedefinieerd als omgeving dat dieren met verhoogde kans op sociale interactie, motorische activiteit en sensorische stimulatie groter dan gebruikelijk ervaren in een standaard laboratorium environment.EE voorzien is aangetoond consequent invloed op het gedrag van dieren, totstandbrenging veranderingen, zoals vermindering van stress en angst-gerelateerde activiteit 8-10, verbeterde prestaties in leer- en geheugentaken 8,11, vroeg begin van de motorische coördinatie en verkennende activiteit 11, veranderingen in de moederlijke zorg 8 evenals de weerstand tegen verslavende stoffen 12- 15. Verder is EE geopenbaard om de effecten van neurodegeneratieve aandoeningen verbeteren, vertragen van het begin en het verminderen van de ernst van de symptomen bij diermodellen van Huntington 1-4,16, Parkinson 17 en ziekte van Alzheimer 18.
Deze veranderingen corverband met de anatomische en moleculaire veranderingen EE bekend induceren gehele hersenen. Dieren die in verrijkte omgevingen, van de vroege stadia van ontwikkeling te laten zien een groot aantal neurale veranderingen, zoals toegenomen gewicht hersenen en corticale dikte, dendritische vertakkingen 9,2-22 en synaptische dichtheid 23. EE zowel de omvang en het tijdstip van groeifactor expressie 9,24-30, waarvan is aangetoond bijdragen aan versnelde ontwikkeling van zintuiglijke 25,26,28,29, geheugensteuntje 30, alsook motorische circuits 31,32 veranderen.
Vorige werk heeft onthuld soms tegenstrijdige bevindingen bij het onderzoek naar de invloed van EE, zonder rekening te houden met de verschillende soorten dieren en omgevingen gebruikt worden binnen de afzonderlijke studies 9,24,27,30. Momenteel is er geen consistente en eenvoudige gedragstaak die kan worden gebruikt om de effectiviteit van verschillende EE paradigma verschillende stammen en spec metenies van de dieren.
De Puzzle Box taak is ontworpen als een eenvoudige test om te bepalen inheemse probleem van een dier oplossend vermogen 7. Dieren geplaatst in de open ruimte zijn verplicht om te verwijderen belemmerende materialen gelegen in een kleine opening om toegang te krijgen tot een overdekte regio / onderdak. Elk vak wordt gegeven drie onderzoeken met dezelfde obstructie om drie verschillende cognitieve eigenschappen te beoordelen. De eerste proef levert een basislijn indicatie van inherente of inheemse probleemoplossend vermogen. De tweede proef, op dezelfde dag, geeft een indicatie van het vermogen van het dier te verbeteren en dus versterken strategieën voor het verwijderen van de specifieke obstructie. De derde proef, uitgevoerd op de volgende dag, inzicht in het vermogen van de patiënt te behouden en herinneren de geleerde oplossing van de taak.
De motivatie voor het oplossen van deze "obstructie puzzels" door de dieren kan worden gevarieerd, potentieeloproepen van een aangeboren verlangen om open velden vermijden en schuilen, evenals een inherent rijden hun omgeving 6,7 verkennen. De talrijke potentiële gedrags onderliggende factoren de wens om de puzzel Box lossen stelt verschillende gebieden van de hersenen zijn betrokken bij de bemiddeling taken. Eerder werk heeft aangetoond dat in muriene modellen van schizofrenie, de prefrontale cortex en de hippocampus betrokken zijn bij het verwerven van deze taak 5. Een laesie bij ratten bleek ook een groot aantal hersengebieden betrokken in Puzzel Box prestaties, waaronder diverse thalamuskernen, de hypothalamus, het cerebellum en limbische structuren. Samen vormen deze bevindingen wijzen erop dat de uitoefening van dit probleem oplossen taak behelst een gastheer van neurale structuren in verband met cognitieve functie.
De puzzel doos is met succes gebruikt om het probleem oplossend vermogen van muizen, alsook cognitieve gebreken vertoont door m beoordelenurine modellen van schizofrenie 5-7. Prestaties op de taak is aangetoond dat het zeer consistent te zijn, correleren met resultaten van andere cognitieve gedragstesten 6. Het doel van dit werk was dus aan de Puzzle Box taak aan te passen aan een eenvoudige en betrouwbare middel van het vaststellen van de effectiviteit van EE geworden.
De gepresenteerde gegevens tonen aan dat de puzzel box effectief kan worden gebruikt om het effect van EE beoordelen. Muizen getogen in verrijkte omgevingen consequent nam aanzienlijk minder tijd om obstructie puzzels op te lossen binnen dit gedrags test dan dieren die binnen de standaard laboratorium omstandigheden deed.
