Herein, we describe a colorimetric assay to detect the presence of reducing sugars during bacterial spore germination.
The process of endospore germination in Clostridium difficile, and other Clostridia, increasingly is being found to differ from the model spore-forming bacterium, Bacillus subtilis. Germination is triggered by small molecule germinants and occurs without the need for macromolecular synthesis. Though differences exist between the mechanisms of spore germination in species of Bacillus and Clostridium, a common requirement is the hydrolysis of the peptidoglycan-like cortex which allows the spore core to swell and rehydrate. After rehydration, metabolism can begin and this, eventually, leads to outgrowth of a vegetative cell. The detection of hydrolyzed cortex fragments during spore germination can be difficult and the modifications to the previously described assays can be confusing or difficult to reproduce. Thus, based on our recent report using this assay, we detail a step-by-step protocol for the colorimetric detection of cortex fragments during bacterial spore germination.
Endospores metabolisch sluimerende vormen van bacteriën die het mogelijk maken bacteriën te volharden in ongunstige omgevingen. In veel sporenvormende bacteriën wordt spore vorming geïnduceerd door ontbering nutriënten maar dit proces kan worden geregeld door veranderingen in pH, blootstelling aan zuurstof of andere belastingen 1. Terwijl in hun metabolisch slapende spore vorm, bacteriën verzetten tegen UV-straling, uitdroging, hogere temperaturen en bevriezing 2. Het grootste deel van de kennis over de sporulatie proces komt uit studies in het model organisme, Bacillus subtilis. De sporulatie begint met DNA replicatie en de vorming van een axiaal filament 3,4. Een asymmetrisch septum verdeelt vervolgens de cel in twee ongelijke grootte compartimenten. Hoe groter compartiment, de moeder cel, overspoelt de kleinere compartiment, de forespore. Zowel de moeder cel en forespore coördineren genexpressie aan de spore en de moeder cel uiteindelijk lyseert rijpen, het loslaten van de dormant spore deze in het milieu 1.
De spore structuur en samenstelling is geconserveerd in vele soorten bacteriën. De spore kern heeft een lager watergehalte dan de vegetatieve cel en is rijk aan dipicolinezuur (DPA) 1,2. Tijdens spore vorming, is DPA gepompt in de spore kern in ruil voor water. Rondom de kern is een innerlijke spore membraan, waar in de meeste sporenvormende bacteriën, de receptoren die de kleine moleculen die de kieming (germinants) te stimuleren te herkennen zijn gelegen op 2. Gelegen net buiten binnenste membraan van de spore is een laag van celwand peptidoglycan. Een gespecialiseerde peptidoglycaanlaag (cortex) omgeeft de celwand peptidoglycan en bestaat uit veel van dezelfde componenten als celwand peptidoglycan [alternerende N-acetylglucosamine (NAG) en N-acetylmuraminezuur (NAM)]. Echter, circa 50% van de NAM residuen in de cortex omgezet naar muramic-δ-lactam 5,6 </sup>. Tijdens sporevorming worden deze muramic-δ-lactam residuen die door de spore cortex lytische enzymen (SCLEs), waardoor de cortex worden afgebroken (een proces dat cruciaal is kieming voltooid), maar niet de celwand. Rondom de cortex is een buitenste membraan en verschillende lagen van manteleiwitten 2.
Beheersing van het proces van kieming is van cruciaal belang voor de sporenvormende bacteriën. Kieming wordt gestart wanneer ontkiemende receptoren interageren met hun respectievelijke germinants 2. In veel sporenvormende bacteriën die germinants zijn aminozuren, suikers, nucleotiden, ionen of combinaties daarvan 2. C. difficile sporevorming wordt geïnitieerd door combinaties van bepaalde galzuren [bv taurocholinezuur (TA)] en aminozuren (bijvoorbeeld glycine) 7-10. Hoewel er verschillen tussen de C. difficile kieming route en de paden bestudeerd in andere sporenvormendebacteriën, zoals B. subtilis, voor alle is de absolute vereiste om de spore cortex degraderen om een vegetatieve cel te groeien van de gekiemde spore 2,8. Cortex afbraak kan door de SCLEs CwlJ / SleB (zoals in B. subtilis) of SLEC (zoals in veel Clostridia). Cortex hydrolyse reduceert de beperking op de celwand en spore. Dit maakt volledige kern rehydratatie, een noodzakelijke stap reactiveren veel van de eiwitten die nodig zijn voor cellulaire metabolisme 2.
Wanneer sporen ontkiemen kan de slapende spore verandert van een fase-heldere toestand naar een geleidelijk donkere toestand en dit proces worden gemeten door een verandering van de optische dichtheid (OD) bij 600 11 nm. Een eerdere rapport suggereert dat veel van deze verandering in OD door het vrijkomen van DPA uit de spore 12. Tijdens onze recente studie hebben we getracht de timing van C. vergelijken difficile spore kieming en bewaakt derelease van DPA en cortex fragmenten 9. Voor deze studie was het kritisch om de vrijlating van cortex fragmenten controleren zij begon te worden vrijgegeven door sporen ontkiemen.
De colorimetrische bepaling hier was gebaseerd op een werkwijze voor het detecteren van suikers met reducerende uiteinden ontwikkeld Ghuysen et al. 13. Omdat andere protocollen zijn beschreven voor de detectie van reducerende suikers of 14 hebben die protocollen 15 gewijzigd, kan de literatuur over dit onderwerp verwarrend. Hier hebben we detail een stap-voor-stap werkwijze voor de colorimetrische detectie van reducerende suikers vrijgemaakt ontkiemt C. difficile sporen. Hoewel deze studie maakt gebruik van C. difficile sporen, de reducerende suikers vrijkomt bij de kieming van sporen van andere sporenvormende bacteriën kunnen worden gedetecteerd met dit protocol 9,16,17.
Bij stimulatie, sporen het kiemproces ondergaan verliezen hun resistentie eigenschappen zonder de noodzaak voor macromoleculaire synthese. Wanneer spore kieming wordt geactiveerd, de spore kern releases DPA in ruil voor water 2. Door de hoge DPA gehalte van de slapende spore, de spore kern onder hoge osmotische druk en de gespecialiseerde peptidoglycaan, cortex, helpt voorkomen dat de kern van zwelling door zich vermoedelijk als een belemmering voor uitbreiding 2.
