يستخدم هذا البروتوكول المراسلات الفلورية وفرز الخلايا لتبسيط التجارب في الماكروفاج وخطوط الخلايا التائية. وتستخدم اثنين من البلازميدات لهذه التجارب المبسطة في طرق، وهي CRISPR/Cas9- وDsRed2 التعبير عن البلازميد وplasmid المانحة إعادة التركيب متجانسة التعبير عن EBFP2، والتي يتم دمجها بشكل دائم في مكان Rosa26 في الخلايا المناعية.
