We report on a smart application of carbon nanotubes for kinetic stabilization of lipid particles that contain self-assembled nanostructures in their cores. The preparation of lipid particles requires rather low concentrations of carbon nanotubes permitting their use in biomedical applications such as drug delivery.
We presenteren een gemakkelijke methode om nanostructuur lipide deeltjes gestabiliseerd door koolstof nanobuisjes (CNT) voor te bereiden. Enkelwandige (oorspronkelijk) en meerwandige (gefunctionaliseerde) CNTs worden gebruikt als stabilisatoren Pickering soort olie-in-water (O / W) emulsies. Lipiden namelijk Dimodan U en Phytantriol worden gebruikt als emulgatoren, die boven water zelfassemblage in de bicontinue Pn3m kubische fase. Deze zeer viskeuze fase wordt gefragmenteerd in kleinere deeltjes met behulp van een sonde ultrasonicator in aanwezigheid van conventionele oppervlakte-actieve stabilisatoren of CNT zoals hier gedaan. Aanvankelijk CNTs (poedervorm) worden gedispergeerd in water, gevolgd door verdere ultrasone trillingen met het gesmolten lipide aan de uiteindelijke emulsie. Tijdens dit proces krijgen de CNTs bekleed met lipide moleculen, die op hun beurt worden verondersteld het vetdruppels omringen een fijnverdeelde emulsie die stabiel is maandenlang vormen. De gemiddelde grootte van CNT-gestabiliseerde lipide nanogestructureerde deeltjes in het submicron range, die goed vergeleken met de deeltjes gestabiliseerde toepassing van gebruikelijke oppervlakteactieve stoffen. Kleine hoek röntgen verstrooiing gegevens bevestigen de handhaving van hetzelfde Pn3m kubische fase in de CNT-gestabiliseerde dispersies lipide in vergelijking met de zuivere lipidefase (bulk state). Blauwverschuiving en verlaging van de intensiteiten karakteristieke G en G 'banden CNT waargenomen Raman spectroscopie karakteriseren van de interactie tussen CNT oppervlak en lipide moleculen. Deze resultaten suggereren dat de interacties tussen de CNTs en lipiden zijn verantwoordelijk voor hun onderlinge stabiliteit in waterige oplossingen. Aangezien de concentraties van CNTs toegepast voor stabilisatie zeer laag en lipidemoleculen kunnen de CNTs functionaliseren, wordt de toxiciteit van CNTs waarschijnlijk gering, terwijl hun biocompatibiliteit sterk wordt verbeterd. Vandaar de huidige aanpak vindt een groot potentieel in diverse biomedische toepassingen, bijvoorbeeld voor de ontwikkeling van hybride Nanodrager systemen voor de levering van multiple functionele moleculen als in combinatietherapie of polytherapie.
In de afgelopen decennia heeft de nanotechnologie ontpopt als een krachtig instrument in het bijzonder op het gebied van preklinische ontwikkeling van geneesmiddelen ter bestrijding van beruchte ziekten zoals kanker 1. In dit verband nanoschaal structuren met grootte <1000 nm schaal worden onderzocht als transportmiddel verschillende actieve biomoleculen zoals geneesmiddelen, eiwitten, nucleïnezuren, genen en diagnostische beeldvormingsmiddelen 1-4. Deze biomoleculen zijn ofwel ingekapseld in de nanodeeltjes geconjugeerd of op het oppervlak van de nanodeeltjes en wordt vrijgegeven op de plaats van werking door triggers zoals pH of temperatuur 5,6. Hoewel zeer klein in omvang, het grote oppervlak van deze nanodeeltjes blijkt zeer voordelig voor gerichte afgifte van actieve biomoleculen zijn. De controle over de grootte en biocompatibiliteit deeltje van groot belang om de therapeutische werkzaamheid en daarmee de toepassing van nanodeeltjes 7,8 optimaliseren.9-13 lipiden, polymeren 14,15, 16,17 metalen en koolstof nanobuisjes 18,19 zijn gewoonlijk gebruikt als nanocarriers voor verschillende biomedische en farmaceutische toepassingen.
Bovendien Nanodrager toepassingen op basis van lipide zelf-geassembleerde nanostructuren hebben een grote betekenis in veel andere disciplines, met inbegrip van voedsel en cosmetische industrie 20,21. Zo worden ze in eiwitkristallisatie 22, 23 scheiding van biomoleculen, zoals voedsel stabilisatoren bijvoorbeeld in desserts 24 en in de afgifte van actieve moleculen zoals voedingsstoffen, smaakstoffen en parfums 25-31. Zelf-assemblerende nanostructuren lipide niet alleen de mogelijkheid om biologisch actieve moleculen in gecontroleerde en doelgericht 32-38, maar ze kunnen ook de functionele moleculen te beschermen tegen chemische en enzymatische afbraak 39,40. Hoewel planaire fluïdum bilaag de commop nanostructuur gevormd door amfifiele lipide moleculen in aanwezigheid van water, worden andere structuren zoals hexagonale en vierkante ook vaak waargenomen 20,41,42. Het type nanostructuur gevormd afhankelijk van de lipiden moleculaire vormstructuur, de lipidesamenstelling in water als op de fysisch-chemische omstandigheden zoals toegepast als temperatuur en druk 43. De toepassing van niet-planaire lipide nanostructuren bijzonder van kubische fasen, is beperkt vanwege hun hoge viscositeit en niet-homogene consistentie domein. Deze problemen worden opgelost door het dispergeren van het lipide nanostructuren in grote hoeveelheid water olie-in-water (O / W) emulsies die micron of submicron grootte lipide deeltjes. Op deze wijze kan een geschikt product met lage viscositeit bereid worden met behoud van de oorspronkelijke lipide zelf samengestelde structuur binnen de gedispergeerde deeltjes. De vorming van deze intern zelf-geassembleerde deeltjes (afgekort als ISAsomes 44 </sup> Bijvoorbeeld cubosomes van kubische fasen en hexosomes van hexagonale fasen) vereist gewoonlijk een combinatie van een hoog energieverbruik stap en de toevoeging van stabilisatoren zoals surfactanten of polymeren. Recent onderzoek in deze richting toont de toepassing van verschillende vaste deeltjes 45 zoals silica nanopartikels 46, klei 47-49 en koolstofnanobuizen 50 voor het stabiliseren van genoemde emulsies, geschikt aangeduid als Pickering 51 of Ramsden Pickering-emulsies 52.
In de afgelopen jaren, koolstofhoudende nanostructuren zoals enkelwandige koolstofnanobuizen (SWCNTs) zijn meerwandige koolstofnanobuizen (MWCNTs) en fullerenen veel aandacht gekregen als nieuwe biomaterialen 53,54. De belangrijkste aandachtspunten zijn hun toxiciteit 55-58, onoplosbaarheid in water 59 en daarmee hun biocompatibiliteit 56. Een efficiënte manier om deze problemen aan te pakken is het oppervlak functiealization met niet giftige en biologisch verenigbare moleculen zoals lipiden. In aanwezigheid van water, lipiden interactie met CNT's op een manier die hydrofoob oppervlak van CNTs wordt afgeschermd van polaire waterig medium terwijl de lipide hydrofiele kopgroepen helpen hun oplosbaarheid of dispersie in water 60,61. Lipiden zijn integrale bestanddelen van cellulaire organellen en sommige voedselmaterialen derhalve hun versiering idealiter verminderen de in vivo toxiciteit van CNTs. Biomedische toepassingen onafhankelijk van elkaar op basis van CNTs 18,19 en lipide nanostructuren 13/09 zijn in een uitgebreide ontwikkeling, maar de toepassingen die eigenschappen van de twee te combineren zijn nog niet goed onderzocht.
In dit werk, gebruiken we twee verschillende soorten lipiden en drie soorten CNTs waarvan SWCNTs in de oorspronkelijke vorm dat MWCNTs gefunctionaliseerd met hydroxyl- en carboxylgroepen. We hebben zeer lage concentraties CNTs gebruikt om de dispersies te bereiden waarvanstabiliteit hangt af van verschillende factoren zoals het type lipide, type CNT, verhouding lipide CNT gebruikt, alsmede de parameters zoals sonicatie toegepast als voeding en duur. Deze video protocol voorziet in technische details van een methode voor het stabiliseren van kinetisch lipide nanodeeltjes met behulp van verschillende CNT-stabilisatoren.
Stabilisatie van lipidendeeltjes
Drie verschillende CNTs worden gebruikt om de lipide dispersies te stabiliseren; waarvan twee meerwandig en gefunctionaliseerde gebruik -OH en -COOH groepen, en een is enkelwandig en niet-gefunctionaliseerde (oorspronkelijk). De CNTs varieerden in grootte als volgt (diameter x lengte): MWCNT-COOH: 9,5 nm x 1,5 micrometer; MWCNT-OH: 8-15 nm x 50 micrometer; SWCNT: 1-2 nm x 1-3 urn.
